本研究提取、纯化了具有较好生物学活性的猪血小板衍化生长因子(pPDGF),并对其理化特性进行了研究。并发现pPDGF可调节人血管内皮细胞、兔血管平滑肌细胞和人血管成纤维细胞增殖周期, 促进血管组织成分细胞DNA、胶原及钙调素含量的增加。pPDGF可调节鼠胸主动脉的血管舒缩活动, 为血管非内皮依赖性。PDGFβ受体和e-sis, c-fos基因的过盛表达与冠状动脉血管中膜发生增殖或粥样硬化的发生有关。
Abstract:
In the present study, a simple and effective method for purifying porcine platelet-derived growth factor (pPDGF)with a potent biological activity was established and the physicochemical characteristics of pPDGF investigated.The results have revealed that pPDGF can regulate the growth cycle of human umbilical vein endothelial cells, rabbit vascular smooth muscle cells and human vascular fibroblasts, promote DNA and collagen synthesis and content of calmodulin in cells of some vascular tissues.pPDGF can also regulate the motion of the aorta of the rat in a non-endothelium-dependent manner.Overexpression of PDGF beta receptors and csis, c-fos oncogenes is associated with proliferation or sclerosis at the tunica media of the coronary arteries.pPDGF-induced cell migration and proliferation, metabolic disturbances, vasoconstriction, some oncogenes such as c-sis and c-fos overexpression are closely associated with the occurrence and development of diseases related to vascular lesions.
