摘要:
目的 探讨绞股蓝总皂甙(GPS)对注射内毒素的家兔血流动力学 和NO的影响。方法 动物随机分3组 对照组,内毒素(LPS)组及GPS +LPS组。动物静脉 麻醉后,取血测量NO水平;通过二道生理记录仪和SMUP-A型生物信号处理仪测量血流动力 学指标。动物处死后,取血再次测量NO水平。结果 对照组实验前 后各指标没有明显的变化(P>0.05)。LPS组平均动脉血压(MAP)、心率、心室内压峰 值、±dp/dtmax和心力环面积在实验后2 h时没有出现明显的变化,但在实 验后4 h、6 h时明显低于实验前和对照组(P<0.01),实验后NO-2明显高于实 验前。GPS组实验后2 h,4 h时MAP、心率、心室内压峰值、±dp/dtmax和心 力环面积与实验前 及对照组比较,没有明显差异(P>0.05),但在4 h时明显高于LPS组(P<0.01);实验 后6 h时,MAP、 心率、心室内压峰值、±dp/dtmax和心力环面积低于实验前和对照组(P <0.05),但高于 LPS组(P<0.05);实验后的NO-2浓度高于实验前和对照组(P<0.05),但低于L PS组实验后的NO-2浓度(P<0.05)。结论 绞股蓝总皂甙对注射内毒素的家兔血流动力学有保护作用 和抑制一氧化氮大量释放的作用。
Abstract:
AIM To investigate the effects of Gypenosides(GPS) on hem odyn amics and nitric oxide levels in rabbits injected with endotoxin. METHODS[ WT] Rabbits were randomly divided into three groups: control, endotoxin (LPS), and LPS plus GPS. The animals were anaethetized with intravenous injecti on of 20% ethyl urethane. Nitric oxide levels were determined by colorimetric a nalysis.The parameters of hemodynamics, including mean arterial pressure (MAP), heart rate (HR), peak value of ventricle pressure (PEAK), ±dp/dtmax , area of cardiac force circle (AREA), peaktime (PT), left ventricle end diastolic pre ssure (LVEDP) and time duration of ascendent and descendent slope (±T), were me asured by SMUP-A, a bioinformatic processor. RESULTS All the investig a ted values in the control group were not significantly changed during the experi ment. Compared with those of the LPS group, the values of HR, PEAK, ±dp/d tmax and AREA, PT, LVEDP and ±T changed less in GPS group at 4 h and 6 h after the administration of GPS. The plasma level of nitric oxide in the GPS group was lower than that in the LPS group but higher than that in the control group after the administration of GPS. MAP showed significant difference between the LPS and GPS groups, and both of them w ere lower than that of the control group. The investigation demonstrated that Ge penosides prominently improved the hemodynamics of the rabbits, and delayed the animal death caused by endotoxin. CONCLUSON GPS might protecte endotoxin-ind uced impairment of hemodynamics in rabbits, that initiation and development of endotoxin- induced shock might be retarded by administration of GPS,and that the mechanism of the improvement might involve the decrease of NO lev el and the improvement of hemodynamics.
