摘要:
目的 观寨lL—l8对基础状态及精氨酸加压索(AVP)诱导心肌或纤维细胞(CFs)增殖和p27蛋白表达的彩响。方法 录用胰酶消化，整逮贴壁法培养CFs，四氟唑盐(MTT)比色法检测细胞增殖，碘化两啶(PI)标记细胞DNA，间接免疫组化染色标记细胞内的p27蛋白，用流式绍胞分析仪(FCM)技术测定细胞周期和p27蛋白表达的阳性率．站果 ①I×10 mol／LAVP组M1vr法测定的CFs的吸收(A)值(0．386士0 011)较基础状态(0．324士0007~)明显丹高(P<0．01)；1×lO U／LIL-坤单独作用组(0．298+0．030)和AVP+IL-I[t组(O．332士O．O4L)的A值均分别较基础状态和AVP组显著降低(均P
Abstract:
AIM To study the effect of IL一16on cardiac fibroblasts(CFs)proliferation and p27 expression under basal condition and induced by arginine vasopressin(AVP)。METHODS Neonatal rat CFs was cultured with enzymatic dissociation．MTT assay was used to measure cell prolireation．I abled with PI，FITC，respectively。DNA 1ahled with PI and p27 expression were deter．mined through flow cytometre(FCM)assay．RESULTS ①A value of CFs measured with MTTassay in basal condition and AVP group were 0．324士0．007 and 0．386士0．011，respecdvely，which were significantly higher than that in II -18 group and AVP+11J一1p group．@Percentage of S stage cell and proliferation index(PI)in IL-I~group and AVP+II -lB group were significantly Iower than those in basal and AVP group．respectively。with commitment of an apparent increase of G0／G1 stage percent(both P<0．01)．⑤Positive rate of p27 expression in II 一1口group was (95．03±1．02)％ ，which was higher than that of basal condition group(78．45士1．19) (P<0.01)．Moreover,positive rate of p27 in AVP+II一-1B group (88.23士2.87) was also higher than that in AVP group(63．30士1．85) (P<0．01)．CONCLUSION IL-13 play an important negative role in CFs proliferation by upregulating p27 expression.
