摘要:
目的 探讨精氨酸加压素(AVP)对心肌成纤维细胞(CFs)与胶原构成的凝胶系统收缩的影响和β1整合素反义寡核苷酸的干预作用.方法 设计合成反义、正义和错配β1整合素寡核苷酸片段,建立体外培养的成纤维细胞与胶原构成的凝胶三维培养体系,计算凝胶块的收缩指数。结果 ①AVP浓度≥1×10-7mol/L时,CFs的胶原凝胶收缩指数明显高于对照组(P<0.01);②1×10-7 mol/L AVP作用4 h凝胶收端指数即大于对照组(P<0.01).并可持续48 h以上;③反义β1整合素寡核苷酸与AVP共同作用组的胶原凝胶收缩指数明显低于AVP组、正义链+AVP组和错配链+AVP组,并均有非常显著性意义(P<0.01)。结论 β1整合素反义寡核苷酸具有特异性抑制AVP刺激CFs胶原凝胶收缩的作用。说明β1整合素在AVP促CFs迁移中起介导作用。
Abstract:
AIM To observe the influence of argipressin(AVF)on the collagen gel contraction by cardiac fibroblasts(CFs),and the inteHerence effect of β1 integrin antisense oligonucleotides.METHODS A three-dimensional culture system of cardiac fibroblasts from SD neonataI rats was set up,as well as phosphorothioate modified oligonucleotides and AVP were added to the coltagen gel system,the index of contraction was calculated.
