目的 研究腺苷对大鼠心肌缺血再灌注(I/R) 致心律失常的影响以及与ATP 敏感性钾通道(KATP)的关系。方法 采用结扎大鼠左冠状动脉前降支造成缺血10 min, 然后再灌注15 min, 诱发室性心律失常。在心肌缺血前分别给予腺苷(2, 6mg/kg) 及KATP阻断剂格列本脲(Glib, 7. 5mg/kg)。结果 ①与对照组相比, 腺苷2, 6mg/kg 两剂量组可明显减少I/R 室颤的发生率(P<0.01), 降低心律失常评分分值(P<0.01), 能明显降低磷酸肌酸激酶(CPK)和乳酸脱氢酶(LDH) 活性(P<0.01)。②Glib 阻断KATP后, 诱发出严重的室性心律失常, 且CPK,LDH 值均显著高于对照组(P<0.01)。③腺苷6 mg/kg 在Glib 阻断KATP后, 亦可降低室性心律失常分值, 降低CPK,LDH 水平, 同Glib组相比较, P<0.01 或P<0.05。结论 腺苷对大鼠I?R 心肌损伤有明显的保护作用, 而KATP的开放可能是其保护作用的部分机制。
Abstract:
AIM To study the protective effects of adenosone (Ado) on ischemic and reperfusive arrhythmias in rat and to probe the role of KATP in its effects. METHODS Left anterior descending artery of the rats were ligated with ischemia for 10 min, and then relaxed to be reperfused for 15 min to observe the effects of A do and Glibenclamide (Glib ). RESULTS ①Compared with control group , the two dose groups of intravenous Ado (2, 6 mg/kg) decreased significantly the incidence of ventricular fibrillation (VF) and arrhythmia scores (P<0. 01), and decreased the serum creatine phosphokinase (CPK) and lactate dehydrogenese (LDH ) (P<0.01). ②Glib orally inhibited the ATP-sensitive potassium current (KATP) , induced more sever ventricu ar arrhythmia, and significantly increased the serum level of CPK and LDH compared with control (P<0.01). ③ The antiarrhythmia effects of the small dose of Ado (2mg/kg) were reversed by Glib, but the large dose (6 mg/kg) remained partially the protective effect on the ischemia and reperfusion injury in rats. CONCLUSION Ado had significantly protective effects on the ischemia and reperfusion injury in rats, and the activation of KATP might be involved in its mechanism.
