摘要:
目的 观察肾上腺素(AD) 或去甲肾上腺素(NA) 刺激体外培养的大鼠心肌细胞凋亡, 探讨其作用机制。方法 大鼠心肌细胞分别暴露于10- 7mol/L 的NA , 10- 7mol/L 的AD,NA + 酚妥拉明(10- 7mol/L ) ,NA + 艾司洛尔(10- 7 mol/L ) ,AD+ 酚妥拉明(10- 7mol/L ) 或AD+ 艾司洛尔(10- 7mol/L ) 24 h。流式细胞仪检测凋亡水平。结果 NA 使凋亡细胞百分率由(4. 2±1. 6)% 增加到(10. 0±5. 0)% (P < 0. 01; n= 8) , 此作用可被艾司洛尔完全抑制, 而不受酚妥拉明影响。AD 与NA 的作用相仿, 使凋亡细胞百分率增加到(12. 8±7. 9)% , 且AD 诱导的凋亡同样被艾司洛尔完全抑制但不受酚妥拉明的影响。结论 AD 和NA 可能通过激活B肾上腺素能通路, 促进体外培养的大鼠心肌细胞凋亡。
Abstract:
AIM To determine whether adrenaline (AD ) or no radrenaline (NA ) can stimulate cardiac myocyte apoptosis in vitro, and it’s possible mechanism. METHODS Rat myocytes were exposed to NA (10- 7mol/L ) ,AD (10- 7mol/L ) ,NA + phen to lamine (10- 7mo l?L ) ,NA + esmo lo l (10- 7 mo l?L ) , AD + phentolamine (10- 7mol/L ) and AD + esmolol (10- 7mol/L ) for 24 h respectively. The levels of apoptotic cells were measured by flow cytometry. RESULTS NA increased the percentage of apoptotic cells from (4. 2±1. 6)% to (10. 0±5. 0)% (P < 0. 01, n= 8) , and this effect was abolished by esmolol but was not affected by phentolamine. The effect of AD was similar to that of NA ,AD increased the percentage of apoptotic cells to (12. 8±7. 9)% and was also abolished by esmolol but not by phentolamine. CONCLUSION NA and AD stimulate cardiac myocytes apoptosis in rat in vitro, via the B-adrenergic phthway.
