摘要:
目的 探讨纳络酮(naloxone,NAL ) 对失血致低血容量性休克大鼠的治疗作用。方法 颈总动脉放血, 使平均动脉压(MAP) 降至6.0 kPa,稳定1h, 完成失血致血容量性休克大鼠模型,21只大鼠随机均分为: 休克组、纳络酮治疗组、正常对照组。观察NAL治疗前后及各时相点大鼠呼吸频率(RR)、MAP、血液指标等各参数的变化。结果 纳络酮组给药后,MAP 即刻升高, 5min 后MAP升至(13.6±2.0)kPa,较休克组增高明显(P<0.01),休克组、纳络酮治疗组失血后RR 明显减慢, 纳络酮给药后30minRR 升至高峰(84±10)/min, 与休克组相比明显增加(P<0.01)。NAL降低了肺体指数和肺含水率; 升高pH, PaCO2, 降低PaCO2, 改善酸中毒和心肺功能。结论 纳络酮在失血致低血容量性休克大鼠治疗过程中有重要作用, 能有效减轻休克的严重程度。
Abstract:
AIM To study the curative effect of naloxone (NAL ) on hemorrhagic shock of rats.METHODS Hemorrhagic shock was performed in Wistar rats by carotid artery bleeding to maintain the mean arterial pressure (MAP) at 45 mm Hg (6. 0 kPa) for one hour. Twenty-one Wistar rats were randomly divided into three groups. group 1 (NS, n = 7) , served as normal controls; group 2 (HS, n = 7) , as hemorrhagic shock and group 3 (NAL , n = 7) , as NAL + HS treatment. Blood gases, mean arterial pressure, respiratory rhythm , lung/ body weight index(LBI) , and rate of lung water (RLW ) were determined. RESULTS 5 min after treatment with NAL , the MAP increased to (13. 6±2. 0) kPa, as compared with that of the HS group (P < 0. 01). In both the HS and the NAL groups, respiratory rhythm reduced following hemorrhage, but it increased in the NAL group and reached the peak on 30th min after the treatment (84±10)min (P < 0. 01). Morveover, it was found that NAL markedly increased blood pH and PaO2 vales,reduced PaCO2 and LBI and RLW , and improved acid-base imbalance in rats following shock.CONCLUSION NAL plays an important role in the treatment of hemorrhagic shock, as it can effectively attenuate the severity of shock.
