摘要:
目的 研究非心脏缺血预处理对未成熟心肌和心肌间质的保护作用。方法 建立动物离体心脏Langendorff灌注模型,分为4组:①缺血/再灌注(I/R,n=6),离体心灌注15 min转为工作心15 min;②心脏缺血预处理组(IPC,n=6),离体灌注15 min转为工作心15 min,缺血5 min/再灌5 min各2次;③双下肢缺血预处理组(DL-IPC,n=6),反复3次阻断双下肢血流5 min/松开5 min,建立离体心脏Langendorff灌注模型,灌注15 min转为工作心15min;④肾缺血预处理组(K-IPC,n=6),反复3次阻断左肾动脉5 min/放开5 min,建立离体心脏Langendorff灌注模型,灌注15 min转为工作心15 min。然后各组全心缺血45 min,恢复灌注15 min改为工作心30 min。以左室功能恢复、心肌含水量、血清肌酸激酶(CK)和乳酸脱氢酶(LDH)漏出率、心肌组织ATP和丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性、心肌羟脯氨酸(HP)含量及血清内皮素(ET)含量作为观察指标。结果 IPC,DL-IPC及K-IPC组左室功能恢复优于I/R组(P<0.05),ATP含量、SOD活性、HP含量均优于I/R组(P<0.01),心肌含水量低于I/R组(P<0.05),MDA含量,CK,LDH漏出率及ET含量均低于I/R组(P<0.01)。结论 非心脏缺血预处理与心脏缺血预处理对未成熟心肌和心肌间质具有明显保护作用。
Abstract:
AIM To determine the protection effects of non-myocardial ischemic precoditioning on immature myocardium and myocardial interstitium .METHODS Isolated perfused neonatal rabbit hearts from group ischemic-reperfucion(I/R),group ischemic preconditioning(IPC),group double limb-ischemic preconditioning (DL-IPC) and group kidney-ischemic preconditioning(KIPC) were underwent 45 minute ischemic followed 45 minute reperfusion before group I/R no IPC,group IPC with 2×IPC(5 minute ischemia followed 5 minute reperfusion),group DL-IPC with 3×IPC(double limbs were obstructed 5 minute followed 5 minute reperfusion). The K-IPC with 3×IPC(right kidney artery was clamped 5 minute followed 5 minute reperfusion). The left ventricular function recovery,MWC,LDH and CK leakage,MDA and ATP content and SOD activity HP and ET content were tested.RESULTS The recovery of postischemic heart function,ATP content,SOD activity and HP content in group IPC,DL-IPC and K-IPC were higher than that of group I/R. MWC,MDA and ET content,LDHand CK leakage in group IPC,DL-IPC and K-IPC were lower than that of group I/R.CONSLUCION DL-IPC and K-IPC had the same cardioprotection to immature myocardium and myocardial interstitium as IPC.
