摘要:
目的 探讨血管钠肽(VNP) 对肺动脉平滑肌细胞(PASMC) 增殖及钠尿肽B 受体(NPR-B) 基因表达的影响。方法 体外培养大鼠PASMC, 分别用噻唑蓝(MTT) 比色法、总蛋白含量测定、放免分析及打点杂交的方法, 观察了VNP 作用下PASMC 的MTT 吸光值、总蛋白含量、细胞内cGMP、cAMP 浓度和NPR-B 的mRNA 水平。结果 VNP 可以显著抑制血清刺激的PASMC 增殖, 升高细胞内cGMP 浓度及NPR-BmRNA 水平, 但对细胞内cAMP浓度无明显影响。结论 VNP 对PASMC 增殖及NPR-B 基因表达具有调控作用, 并且可能通过cGMP 途径发挥上述作用。
Abstract:
AIM To investigate the effect and mechanism of vasonatrin peptide (VNP) on the proliferation of pulmonary artery smooth muscle cell (PASMC) and gene expression of natriuretic peptide B receptor (NPR-B), METHODS The optical density, protein content, intracellular concentration of cGMP, cAMP and mRNA level of NPR-B were measured by means of MTT colorimetry, protein colorimetry, radioimmunological analysis and dot hybridization respectively. RESULTS VNP significantly inhibited the proliferation of PASMC stimulated by serum,elevated the concentration of cGMP and mRNA level of NPR-B, but had no significant effect on the level of cAMP.CONCLUSION VNP may regulate the proliferation and expression of NPR-B in a cGMP dependent manner.
