摘要:
目的 检测猪血小板衍生生长因子(pPDGF)的致丝裂活性。方法 以NIH3T3细胞作为靶细胞,利用3H-TdR掺入合成DNA的方法,检测pPDGF的致丝裂活性。结果 pPDGF可促进处于静止状态的NIH3T3细胞DNA合成,并呈现出明显的浓度依赖关系,在30 ng/ml的浓度时DNA合成达到高峰,24 h DNA合成最为显著,经2-巯基乙醇处理的pPDGF其致丝裂活性基本丧失。结论 pPDGF具有良好的生物学活性,2-巯基乙醇可使其丧失生物学活性。
Abstract:
AIM To detect the mitogenic activity of porcine platelet-derived growth factor (pPDGF).METHODS In the present experiment,the NIH3T3 cells were cultured and the mitogenic activity of pPDGF were investigated with3H-TdR incorporation in cultured NIH 3T3 cells. RESULTS pPDGF could stimulate DNA synthesis of quiescent NIH 3T3 cells in a concentration range of 1~30 ng/ml dose-dependently,with a maximal response at a concentration of 30 ng/ml at 24 h. The mitogenic activity of pPDGF was destroyed with 2-mercaptoethanol.CONCLUSION pPDGF had a strong mitogenic activity,but lost its biological activity with 2-mercaptoethanol.
