摘要:
目的与方法 在儿茶酚胺诱导的大鼠心肌肥厚及培养的心肌细胞中,观察κ-阿片受体选择性激动剂U50,488H对心肌肥厚及心肌细胞生长的作用。结果大鼠静脉注射异丙肾上腺素2周(3 mg·kg-1·d-1)发生心肌肥厚,表现为心脏重量指数明显增加,光镜和电镜显示明确的肥厚性组织病理学改变。U50,488H可明显改善这些异常改变。且对去甲肾上腺素引起的促心肌细胞生长具有明显的抑制作用。结论κ-阿片受体在抑制心肌细胞的生长和心肌肥厚的发生发展中具有重要的负性调节作用。
Abstract:
AIM and METHODS To investigate the effects of U50,488H (a selectiveκ-opioid receptor agonist) on cardiac hypertrophy and cardiac cells growth induced by catecholamine.RESULTS The cardiac hypertrophy was developed following isoproterenol ((3 mg·kg-1·d-1)intravenously for 2 weeks. The cardiac weight index increased significantly with the hypertrophic alteration in light micrographs of cardiac tissue and transmission electron micrographs of myocardial ultrastructure. Administered U50,488Hsignificantly attenuated these abnormal changes and markedly inhibited the cardiac cells growth stimulated by norepinephrine.CONCLUSIONκ-opioid peptide attenuates the cardiac hypertrophy, andκ-opioid receptor plays an important negatively role in the development of cardiac hyperytrophy and cell growth.
