目的:探讨洋葱提取物(Extraction of Onion, ETO)对小鼠的急性毒性和大鼠血压、心电图、心率和呼吸的影响。方法: 对正常大鼠用去甲肾上腺素药物诱导和单侧颈总动脉阻塞造成高血压动物模型, 按45.24 g·kg- 1·d-1ETO (0.58 g·m l- 1×3 次) 给小鼠灌胃(ig); 60.32 g·kg- 1·d- 1 ETO 给小鼠腹腔注射(ip);观测ETO的小鼠急性毒性。以2. 32 g·kg- 1 ETO 给大鼠颈外静脉注射(iv), 用八道生理记录仪分别测量不同时间的血压、心电图、心率等指标。结果: ①ETO ig 小鼠的最大耐受量(MTD)大于45. 24 g·kg- 1; ip 小鼠的MTD大于60.32 g·kg- 1。②ETO 2.32 g·kg- 1对正常大鼠血压具有瞬间明显降血压作用(P<0. 05); ③ETO 2.32 g·kg- 1对药物诱导和阻塞性高血压大鼠的血压具有显著降压作用, 给ETO 后20 s 时平均降低幅度分别为3.48 kPa 和7.53 kPa (P<0. 01) ,且降血压的特点明显不同。前者仅表现为瞬间降压; 而后者则为瞬间血压降低幅度大, 之后回升至接近正常水平并维持120 s。④去甲肾上腺素药物诱导和单侧颈总动脉阻塞, 除显著加快心率和呼吸频率外, 对大鼠心电图无明显影响; ETO 2.32 g·kg- 1 iv 后除有显著降血压作用外, 未发现心电图、心率和呼吸的明显变化。结论: ①ETO 毒性甚小; 小鼠的MDT 大于45.24 g·kg- 1 (ig) 和60.32 g·kg- 1 (ip)。②ETO 对正常血压具有瞬间明显降血压作用。③ETO 2.32 g·kg- 1对去甲肾上腺素和单侧颈总动脉阻塞性高血压大鼠具有显著降血压作用。
Abstract:
AIM: To investigate the acute toxicity of ETO (Extraction of Onion) and its effect on blood pressure, elect rocardiogram (ECG), heart rate and respiration of rats. METHODS: 30 mice were given ETO in a dosage of 45.24 g·kg- 1·d- 1 by ig or 60.32 g·kg- 1·d- 1 by ip and ETO’s acute toxicity on mice was observed. 30 rats were randomly divided into three groups. In the control group, physiological solution was administered by ig. In the noradrenalin group, the dosage of no radrenalin was 1 m g ·kg- 1. In the third group, one side of common carotid artery was blocked. After treated with above measures respectively, the rats were all given ETO in a dose of 2.32 g·kg- 1 by iv. The record was tracked and rats’ blood pressure, ECG and heart rate were measu red at different time points. RESULTS: ① The ETO’s MTDs were more than 45.24 g·kg- 1 by ig and 60.32 g·kg- 1 by ip respectively. ②ETO could instantly lower blood pressure of normal rats (P<0.05). ③ETO 2.32 g·kg- 1 could significantly decrease blood pressure of rats with hypertension induced by medicine and block, and the average extents were 3.48 kPa and 7.53 kPa at the point of 20 s respectively (P<0.01). ④ Noradrenalin and blocking one-side common carotid artery could expedite heart rate and respiration rate and had no effect on ECG. ETO did not cause obvious changes in rats’ ECG, heart rate and respiration. CONCLUSION: ETO’s acute toxicity is very mild. ETO can markedly lower blood pressure of normal and hypertensive rats and may provide good evidence supporting therapeutic effect against hypertension.
