目的:探讨内皮素-1 (ET-1) 在肺缺血/再灌注(I/R) 损伤中的作用。方法: 健康新西兰大白兔30只随机分成3组(每组10只): 假手术组(?组)、I/R + 生理盐水(?组)和I/R + ET-1 抗体组(?组)。观察肺再灌注0,60, 120, 240min各时间点肺动脉压(PA)、动脉氧分压(PaO2)和血浆ET-1的变化。实验结束时, 检测支气管肺泡灌洗液(BALF)中ET-1和肺组织丙二醛(MDA) 的含量和肺组织光镜的变化。结果: ? 组血浆和BALF 的ET-1水平、肺组织MDA 含量和PA 升高, PaO2降低。?组PA和PaO2没有明显变化, 血浆和BALF的ET-1水平、肺组织MDA含量的升高得到抑制。结论: ET-1 参与肺缺血?再灌注损伤, 能改善PA 和PaO2。
Abstract:
AIM: To investigate the effect of endothelin-1 on lung ischemia/reperfusion (I/R) injury in rabbits. METHODS: Thiry New Zealand rabbits were divided into three groups: group ?, sham-operated; group ?, I/R + physiologic saline and group ?, I/R + endothelin-1 antibody. Pulmonary artery pressure (PA) , arterial oxygen tension (PaO2) and plasm a endothelin-1 (ET-1) were measured at different time points. The concent rations of ET-1 in bronchio-alveolar lavage fluid (BALF) and malondialdehyde (MDA) in the lungs were analysed. Mean while, the histopathologicall alterations in the lungs were observed. RESULTS: The marked increase of PA, ET-1, MDA and the decrease of PaO2 were observed after lung ischemia/reperfusion, whereas in the ET-1 antibody PA were significantly devreased and PaO2 was reduced. CONCLUSION: ET-1 might participate in the injury due to the lung ischemia/reperfusion and improve PA and PaO2.
