目的:探讨大鼠心肌缺血后不同再灌注时相心肌细胞凋亡的变化及其与心功能之间的关系。方法:60只大鼠随机分成假手术组,缺血 45min再灌注即刻、再灌注 2、4、8h组。以缺口末端标记法(TUNEL)标记凋亡细胞,左室插管监测左室功能变化情况。结果:心肌细胞凋亡随心肌缺血/再灌注不同时相而变化,其中细胞凋亡指数(AI)于再灌注后 2、4h最高(24±4;20±3);细胞凋亡与血肌酸磷酸肌酶(CK)呈正相关(r=0.92,P<0.05),与左室内压上升最大速率(+dp/dtmax)呈负相关(r=-0.81,P<0.05)。结论:心肌细胞凋亡随再灌注不同时相而变化,细胞凋亡可加重心肌缺血/再灌注损伤,进而加重心功能的损害。
Abstract:
AIM:To study changes in apoptosis and cardiac function with different durations of reperfusion after myocardial ischemia. METHODS: 60 rats were divided randomly into 5 groups, sham-operated (observing for 12h in operated control), 45 min of ischemia followed by 0 h of reperfusion (I 45 min R 0 h), I 45 min R 2 h, I 45 min R 4 h and I 45 min R8 h. The myocardial cell apoptosis was determined with terminal deoxynucleotidyl transferase-mediated dUTP-fluorescein nick end labeling(TUNEL) method. RESULTS: Cardimyocyte apoptosis varied with the duration of reperfusion.The apoptosis was positively correlated to CK(r=0.92,P<0.05),but negatively correlated to ventricular function (r=-0.81,P<0.05).CONCLUSION: The number of apoptatic myocytes varied with the duration of myocardial reperfusion. Changes in apoptosis may be invovled in the process of myocardial ischemia reperfusion injury.
