摘要:
目的:探讨静脉注射常规剂量阿霉素(Adriamycin , ADR或Doxorubicin , DXR) 建立兔心衰动物模型的可行性,为 研究心衰的发病机制和有效治疗措施提供理想的动物模型。方法:日本大耳白兔19 只,随机分成模型组( n = 12) 和 对照组( n = 7) 。模型组:注射ADR(1 mg/ kg ,用0. 9 % NaCl 注射液配制成1. 0 mg/ ml 溶液) ,每周2 次,共8 周;对照 组:注射相同体积0. 9 % NaCl 注射液。于末次注射停药后3 周,测量其体质量(BW) 、心室质量(VW) 、左室收缩压 (LVSP) 、左室舒张末压(LVEDP) 及最大压力上升及下降速度( ±d p/ d tmax) ,并作病理切片观察其组织学变化。结果: 与对照组相比,模型组动物VW/ BW明显增大,LVSP 明显降低,LVEDP 明显升高, ±d p/ d tmax则显著减小( P < 0. 01) , 病理学结果证实符合心肌病样改变。结论: :常规剂量多次静脉注射ADR 可显著损伤心脏的舒缩功能,是建立兔心 肌病及慢性充血性心力衰竭模型的一种简便可靠的方法。
Abstract:
AIM: To study the feasibility of establishment a rabbit of adriamycin-induced heart failure , provide a considerable model for investigation of mechanism and effective therapy in hear failure. METHODS : There are 19 rabbits were randomly divided into 2 groups : The model group ( n = 8) was intravenous injection of ADR at a dose of 1mg/ kg twice weekly for 8 weeks , the control group ( n = 7) received an equivalent volume of 0. 9 % saline alone. 3 weeks after the final injection , body weight (BW) ,ventricular weight (VW) ,left ventricular systolic pressure (LVSP) ,Left ventricular end2diastolic pressure (LVEDP) and ±d p/ d tmax were evaluated. Hearts were harvested after that and analyzed by histological hematoxylin2eosin staining. RESULTS : Compared to the control group , VW/ BW and LVEDP of the model group increased , LVSP and ±d p/ d tmax decreased. Pathological examination demonstrated that the cardiac muscular tissue of the model group were harmed , which characteristic consistent with cardiomyopathy. CONCLUSION: Chronic administration of intravenous ADR to rabbits harmed systolic and diastolic function of hearts ; it is a simple and reliable method to establish a model of cardiomyopathy and chronic congestive heart failure.
