目的: 探讨粒细胞集落刺激因子( G2-CSF) 和干细胞因子(SCF) 预处理对骨髓间充质干细胞(MSCs) 向心肌细 胞分化的影响。方法: MSCs 均采用全骨髓法提取,通过换液传代逐渐纯化细胞,取传4 代的MSCs ,在共培养前用 DAPI 标记,心肌细胞在培养的第3 d 与MSCs 共培养。对照组:未施加任何干预的MSCs 与心肌细胞共培养;实验 组:联合使用G-CSF 和SCF 对大鼠在体动员,提取其MSCs 与心肌细胞共培养。共培养前测定心肌细胞的活力,在 共培养第3 、4 、5 d 用免疫荧光标记肌钙蛋白T , 分析DAPI-MSCs 向心肌细胞分化的百分率。结果: 在共培养的第 3 、4 、5 d ,G-CSF 和SCF 处理的DAPI-MSCs 的分化率分别为(1. 24 ±0. 16) %、(3. 46 ±0. 21) %、(7. 21 ±0. 11) % ,均 显著高于对照组的0 、(1. 27 ±0. 13) %、(1. 32 ±0. 25) %。结论: : G-CSF 和SCF 对MSCs 具有促分化作用。
Abstract:
AIM: To investigate the effect of pret reatment of Granulocyte Colony-Stimulating Factor ( G-CSF) and Stem Cell Factor (SCF) on the differentiation of mesenchymal stem cells(MSCs) into cardiomyogenic cells. METHODS: The MSCs were isolated primarily f rom bone marrow of adult rat , and purified by passage culture. The 4th passage of MSCs was labled by DAPI . Then cardiomyocytes that have been cultured for 3 days were cocultured with DAPI-MSCs . The cont rol group : The MSCs which have no intervention were cocultured with cardiomyocytes. The experiment group : The MSCs which have been pret reated by SCF and G-CSF were cocultured with cardiomyocytes. The activity of cardiomyocytes was detected before coculture. Troponin T was examined by immunocytochemical method in the 3th、4th 、5th day of coculture separately. Then we analyse the percentage of the differentiation of MSCs into cardiomyogenic cells. RESULTS: DAPI-MSCs pret reated by SCF and G-CSF can be induced to differentiate into cardiomyogenic cells with higher positive rate of differentiation[ (1.24 ±0. 16) %、(3. 46 ±0. 21) %、(7. 21 ±0. 11) % in the 3th、4th 、5th day separately ]than the cont rol group[0 、(1. 27 ±0. 13) %、(1. 32 ±0. 25) %] . CONCLUSION: G-CSF and SCF could induce MSCs to differentiate into more cardiomyogenic cells in simulative cardiomyocytes microenvironment s in vitro.
