摘要:
目的: 探讨肾缺血预处理(RIP) 对未成熟心肌细胞凋亡相关蛋白表达的影响。方法: 采用兔Langendorff 模型,幼兔24 只随机分为3 组,对照组(C , n = 8) :仅灌注KH 液180 min ,然后取标本;缺血/ 再灌组( I/ R , n = 8) :灌注20 min 后,停灌45 min ,再灌120 min ;肾缺血预处理组(RIP , n = 8) ,反复2 次阻断肾血流5 min/ 放开5 min ,建立Langendorff 模型,然后重复I/ R 组缺血/ 再灌方法。以凋亡细胞原位标记与半定量分析、琼脂糖凝胶电泳检测DNA 片段梯及Bcl-2 、Bax、Fas 基因蛋白的表达作为观察指标。结果: 凋亡细胞原位标记与半定量分析:RIP 组与I/ R 组比较,心肌细胞凋亡率显著减少;琼脂糖凝胶电泳检测DNA 片段梯: RIP 组与I/ R 组比较,光密度显著降低;RIP 组与I/ R 组比较,Bcl-2 表达明显增多,Bax、Fas 表达显著减少。结论:RIP 可能通过调节Bcl-2 、Bax、Fas 蛋白的不同表达影响心肌细胞的凋亡。
Abstract:
AIM: To investigate the effect s of renal ischemic preconditioning (RIP) on immature my-ocardial cell apoptosis and expressions of Bcl-2 ,Bax and Fas proteins. METHODS: Isolated rabbit heart Langendorff model was used in this study. 24 immature rabbit s (aged 14~21 days) were randomly divided into 3 groups. The cont rol group were only perfused for 180 minutes. The ischemia/ reperfusion group were perfused 20 minutes ,and left with myocardial ischemia for 45 minutes and then followed by reperfusedion for 120 minutes. The right kidney arteries of the RIP group were clamped 5 minutes and then followed by 5 minutes reperfusion 2 times ,and then by repeating the method of I/ R group. The terminal t ransferase U TP nick end labeling ( TUNEL) , nucleosomal ladder of DNA f ragment s , and the expression of Bcl-2 , Bax ,and Fas protein were detected. RESULTS : There were marked reduction in TUNEL cell apoptosis ,ladder of DNA f ragment s ,expressions of Bax and Fas protein ,and marked increase in expression of Bcl-2 protein in the RIP group ,compared with the I/ R group. CONCLUSION:This study demonst rated that RIP decrease immature myocardial cell apoptosis by regulating the expressions of Bcl-2 , Bax ,and Fas protein.
