摘要:
目的 探讨氯沙坦对心脏保护作用与肝细胞生长因子(HGF)的关系。方法 实验动物为自发性高血压大鼠(SHR)36只,分别为6周龄,14周龄和24周龄。分为两组,一组为氯沙坦组(SL),另一组为对照组(SC);以Wistar大鼠为正常对照组(WC),年龄和体质量配比,每组各6只,雌雄配对。SL组每天按30 mg/kg灌胃法喂服,对照组采用灌胃法喂服等量蒸馏水,疗程6周。实验结束后分别测量血压,左室质量,采用M30apoptosense ELISA法测心肌细胞凋亡数量及血浆,组织HGF浓度,心肌胶原容积和血管周围胶原容积。结果 治疗6周后,与未服药的对照组相比,SL组不但血压下降,同时左室质量指数降低,间质纤维化及血管周围纤维化程度减轻,心肌细胞凋亡数量减少,血浆HGF降低,伴有组织HGF浓度升高。结论 氯沙坦能够促进组织HGF分泌,恢复其自身修复能力而逆转心室重构。同时改善内皮功能,维护心肌组织与内皮的稳定性,可能有降低心脑血管病危险性的作用。
Abstract:
AIM To study the relationship of hepatocyte growth factor and cardioprotection of Losartan during left ventricular remodeling in SHR. METHODS 36 SHRs aged 6,14 24 weeks were randomly divided into two groups and treated for 6 weeks as follows: vehiclen (distilled water, n=18), Losaran(30 mg/d, n=18), compared with 18 WKYs aged 6,14,24 weeks. Systolic blood pressure was measured by the tailcuff method each week. After treatment, the rats were killed. Blood samples were collected by decapitation without anesthesia in prechilled tubes and before measurment were stored at -70℃ . The cardiac tissue were promptly removed from the apex and frozen in liquid nitrogen and then stored at -70℃ until use. The plasma and myocardium HGF were determined by ELISA. Cardiomyocyte apoptosis was identified by M30 apoptosenseTM ELISA. The collagen volume fraction(CVF) and perivascular circumferential area(PVCA) were measured by pathological examination. RESULTS ①Systolic blood pressure, left ventricular weight index, CVF and PVCA were all significantly decreased in SHRsLosartan, compared with SHRcontrol(SC) groups. ②Compared with SC groups, plasma HGF were also strongly decreased at 14 and 24 weeks, but the cardiac HGF were increased accordingly at each stage. CONCLUSION Losartan can effectively reverse left ventricular remodeling of SHR and improve endothelial function via inducing tissue HGF production, which is a potential target to decrease cardio–cerebrovascular risk.
