摘要:
目的 比较大鼠、豚鼠和家兔心房肌和心室肌M受体分布密度及与其强阻断剂氚标记二苯羟乙酸奎宁酯(3HQNB)亲和活性的种属差异性。方法 运用放射性配基受体结合法测定不同动物心房肌和心室肌M受体密度及解离常数(Kd)值。结果 ①大鼠心房M受体的分布密度约为心室的3.0倍,家兔心房是心室的4.0倍,而豚鼠心房是心室的1.7倍。②M受体在不同种动物心房肌密度排序:家兔>大鼠>豚鼠。心室肌密度排序:豚鼠>大鼠>家兔。Kd值在心房、心室及种间均无统计学差异。结论 ①M受体在大鼠、豚鼠和家兔心脏的分布密度不仅具有房室间差异性,而且还有种间差异性。②M受体与其阻断剂3HQNB亲和活性的种间差异性不明显。
Abstract:
AIM To investigate the density of Macetylcholine receptors in atrial and ventricular myocardium of rats, guineapig and rabbits. METHODS Using radioligand binding assay, the Mreceptor density and dissociation constant were observed in atrial and ventricular myocardium of above animals. RESULTS ① Atrial myocardial Mreceptors density is 3.0, 4.0 and 1.7 times bigger than ventricular in rats, rabbits and guineapig respectively. ② The density of Mreceptors in atrial myocardium from high to low is: rabbits, rats and guineapig. whereas ventricular myocardium was found to be: guineapig, rats and rabbits. ③ No significant difference were observed in Kd of atrial and ventricular myocardium of rats, rabbits and guineapig. CONCLUSION The density of Mreceptors is not only different in atrial and ventricular myocardium in animals mentioned above, but also difference among species. In addition, the affinity of M receptors was found to be no speciesrelated.
