摘要:
目的 观察呋喃二氢吡啶二羧酸酯(EFDP)对血管紧张素II(AngⅡ)诱导血管平滑肌细胞(VSMC)增殖的影响。方法 采用培养的兔VSMC,应用3HTdR掺入法,观察在AngII促进VSMC增殖过程中,EFDP对VSMC DNA合成的影响及其时间效应。结果 AngII可促进处于静止状态的兔VSMC的 DNA合成,36 h时细胞DNA的合成达到高峰。EFDP对AngII诱导的VSMC增殖有显著的抑制作用,并呈现出明显的浓度依赖关系,随着EFDP浓度的升高,VSMC增殖的抑制率也逐渐增加,36 h时,78.40 μmol/L 的EFDP对VSMC增殖的抑制率为21.37%。结论 EFDP可抑制AngII诱导兔VSMC的增殖,并存在一定的量效依赖关系及时间反应性。
Abstract:
AIM To investigate effects of diethyl2,6diethyl4furnyl, 4dihydropyridine3, 5 dicarboxylate (EFDP) on proliferation of cultured vascular smooth muscle cells(VSMCs) stimulated by angiotension II(Ang II). METHODS The rabbit VSMCs were cultured and the inhibitory effect of EFDP on DNA synthesis in vascular smooth muscle cells stimulated by Ang II were assayed at different times with different concentrations of EFDP by using 3HTdR incorporation in vitro. RESULTS Ang II could significantly promote DNA synthesis in quiescent rabbit VSMCs, with a maximal response at 36th h. EFDP significantly inhibited DNA synthesis of VSMCs in a dosedependent manner, with a maximal response at a concentration of 78.40 μmol/L and at 36th h when the inhibitory rate was 21.37%. CONCLUSION EFDP significantly inhibits DNA synthesis of ribbit VSMCs induced by Ang II in a dose and time dependent manner.
