摘要:
目的 研究经皮穿刺血管内成形术后大鼠腹主动脉血管平滑肌细胞增殖程度与p27蛋白水平的关系,及国产雷帕霉素(Rapamycin)对血管平滑肌细胞增殖的抑制,探讨防治经皮冠状动脉内成形术(PTCA)后再狭窄的有效途径和指标。方法 流式细胞计数法测定p27蛋白、CDK2、cyclin E在大鼠再狭窄血管平滑肌细胞及对照组的表达水平。 结果 雷帕霉素干预后p27蛋白表达水平在PTCA术后各组与对照组相比均有上移,减轻血管狭窄程度(P<0.01)。 结论 p27蛋白在增殖的血管平滑肌细胞中表达水平与狭窄程度负相关,国产雷帕霉素可上调p27蛋白水平从而抑制血管狭窄形成。
Abstract:
AIM To study the relationship between p27kip1 levels and the degree of proliferation of vascular smooth muscle cell(VSMC) in rats following balloon injury, and the interference by Rapamycin. METHODS percutaneous transluminal angioplasty(PTA) was performed on a rat model. The expression of p27kip1 CDK2 and cyclinE were measured by flow cytormetry. Data were analyzed by SPSS. RESULTS Rats receiving Rapamycin before surgery demonstrated elevated p27kip1 level and alleviated stenosis. CONCLUSION p27kip1 level in proliferated VSMC is negatively correlated to proliferated degree. Rapamycin could reduce proliferation of VSMC after balloon injury by increasing the level of p27kip1.
