摘要:
目的 研究益气活血复方对异丙肾上腺素(Iso)所致急性心肌缺血防治作用。方法 用灌胃的方法,给大鼠灌服益气活血复方(2 mg/kg)两周后,皮下注射Iso(5 mg/kg)以诱导急性心肌缺血模型。并测定心肌线粒体中丙二醛(malondialdehyde,MDA)含量,超氧化歧化酶(superoxide dismutase,SOD)及谷胱甘肽过氧化物酶(glutathion peroxidase,GSHPX)活性;心肌组织及血清中乳酸脱氢酶(Lactic dehydrogenase ,LDH )、肌酸激酶(Creatine kinase,CK)含量。结果 急性心肌缺血模型组中,心肌线粒体中MDA含量增加,SOD活性降低及GSHPX活性增加;益气活血复方组大鼠心肌线粒体中MDA含量降低,SOD、GSHPX活性显著增加;心肌组织中LDH、CK含量显著升高,血清CK、LDH含量显著降低。结论 益气活血复方可能是通过降低心肌线粒体中MDA含量,增加SOD、GSHPX活力,减轻氧自由基的损伤。
Abstract:
AIM To study the protective effect of Qiinvigrating and Bloodactivating Prescription (QIBAP) on the injury of myocardial mitochondrion induced by Isoproterenol (Iso). METHODS Rats were fed with QIBAP for two weeks and acute myocardial ischemia was induced by subcutaneous injection of Iso (5 mg/kg). The content of malondialdehyde (MDA), activities of superoxide dismutase (SOD) and glutathion peroxidase (GSHPX) in myocardial mitochondrion were measured. The content of lactic dehydrogenase (LDH) and creatine kinase (CK) were measured in myocardium and serum in different groups. RESULTS In model group, the content of MDA was increased ( P<0.01 vs Control ), SOD activity of was decreased ( P<0.01 vs control ), but the activity of GSHPX was increased (P>0.05 vs Control). Compared with those in model group, the MDA content of mitochondrion was reduced in QIBAP group and the activity of SOD and GSHPX were increased significantly in QIBAP group. The content of LDH and CK were increased significantly in myocardium, but they were reduced significantly in serum ( P<0.01 vs Control ). CONCLUSION QIBAP may attenuate myocardial injury caused by oxygen free radicals by decreasing the content of MDA and increasing the activities of SOD and GSHPX in myocardial mitochondrion.
