目的 完全房室传导阻滞是心脏外科最严重的常见并发症,窦房结细胞移植是治疗思路之一,窦房结细胞的分离、培养难度很大,成功率低。本研究旨在建立一套可靠的窦房结细胞分离、纯化培养技术。方法 新西兰乳兔的窦房结组织进行原代细胞培养,差速贴壁结合5-溴脱氧尿核苷(BrdU)处理。结果 在培养的窦房结细胞中观察到3种不同形态的细胞,即梭形细胞、三角形细胞与不规则形细胞。其中梭形细胞最多,且形态结构与电生理特征符合窦房结起搏细胞的特点,三角形细胞与心房肌三角形细胞特征相似。差速贴壁技术结合BrdU处理的纯化培养中窦房结细胞比例比常规方法高,分别为65%和45%(P<0.01)。结论 用差速贴壁结合BrdU处理的纯化培养可显著提高窦房结梭形细胞的比例,是一种可靠的窦房结细胞培养技术。
Abstract:
AIM To develop a reliable method for primary culture of isolated sinoatrial node cells. METHODS The isolated neonatal rabbit sinoatrial node cells were cultured by differential attachment and treated with 5′-Bromo2′-deoxyuridine. RESULTS Among the cultured cells, we observed cells of three different configurations: spindle cells, triangle cells and irregular cells. The greatest proportion of the observed cells was spindle cells, whose configuration, structure and electrophysiological characters accorded with those of pacemaker cells of sinoatrial node cells. Triangle cells isolated from the sinoatrial node cells had similar characteristics to those of triangle cells of atrial muscles. The proportion of spindle cells increased from 45% in the cells cultured with routine method to 65% by our method. CONCLUSION Differential attachment and BrdUtreatment can significantly increase the proportion of spindle cells and is a reliable method for primary culture of isolated sinoatrial node cells.
