目的 建立大鼠心肌梗死后心力衰竭(心衰)模型,并进行评估,以提高存活率和成功率。 方法 分批对160只大鼠行冠状动脉左前降支结扎手术,同时设48只假手术组,计算死亡率。在4周时行血流动力学检查,检测心率(HR)、左心室舒张末压(LVEDP)、计算左室压力最大上升速率(+dp/dtmax)和左室压力最大下降速率(-dp/dtmax)。结果 4周时模型组心率、+dp/dtmax和-dp/dtmax绝对值均低于假手术组(分别为P<0.05,P<0.01,P<0.01),LVEDP则高于假术组(P<0.01)。随着手术大鼠例数的增多,存活率得到提高(P<0.05)。结论 心梗后4周形成心衰模型,通过采取必要的措施,可以提高动物生存率。
Abstract:
AIM To establish and assess the rat model of heart failure after myocardial infaction (MI) to improve survival rate. METHODS The rat model of MI was made through ligating the anteriordescending coronary artery in 160 rats. The sham group was the same as MI group but without ligating the coronary artery. Hemodynamic parameters including heart rate, left ventricular end diastolic pressure (LVEDP), maximal velocity increase of pressure per second ( +dp/dtmax)and maximal velocity decrease of pressure per second (-dp/dtmax)were observed at 4th week and the mortality was evaluated. RESULTS At 4th week, HR, absolute value of +dp/dtmax and -dp/dtmax in model group were higher than those in sham group (P<0.05, P<0.01 and P<0.01). The survival rate was significantly improved from the first batch of rats to the last (P<0.05). CONCLUSION The heart failure model forms at 4th week after MI. The survival rate can improve with proper measures taken during and after the operation.
