摘要:
目的 观察骨髓单个核细胞(bone marrow mononuclear cells, BMMNCs)移植对大鼠急性心肌梗死(AMI)后心肌细胞凋亡的影响。方法 健康雄性Wistar大鼠30只,随机均分为实验组和对照组。用结扎冠状动脉左前降支的方法建立大鼠AMI模型,将制备的BMMNCs悬液和培养液分别经心外膜下植入实验组和对照组梗死心肌周围。移植术后4周末,检测心肌细胞凋亡情况及Bcl2、Fas、FasL蛋白在心肌细胞中表达水平,并观察梗死区心肌内移植BMMNCs及其周边区组织形态学特点。结果 移植后4周末,实验组心肌细胞凋亡指数显著低于对照组(P<0.05);与对照组相比,实验组Bcl2蛋白吸光值显著升高(P<0.05),Fas、FasL蛋白吸光值显著降低(P<0.05);实验组心肌梗死区内有BrdU标记阳性的BMMNCs存活。结论 同种异体大鼠BMMNCs移植可调节Bcl2、Fas、FasL蛋白表达,抑制AMI后心肌细胞凋亡发生。
Abstract:
AIM To observe the effects of allogeneic bone marrow mononuclear cells (BMMNCs)on rat cardiac myocyte apoptosis after acute myocardial infaretion(AMI). METHODS Thirty male healthy Wistar rats were randomly divided into two groups: control group(n=15)and transplantation group(n=15). As soon as the descending anterior branch of left coronary artery was ligated, DMEMLG medium was injected into the periinfarct area of control group and BMMNCs labeled with BrdU were injected into the periinfarct area of transplantation group. Apoptotic cells and the expression of Bcl2, Fas and FasL were detected 4 weeks later. RESULTS Four weeks after the transplantation, cardiomyocyte apoptotic index and the expression of Fas and FasL decreased significantly(P<0.05) while the expression of Bcl2 increased significantly(P<0.05)in the transplantation group, compared with those in the control group. BMMNCs labeled with BrdU were observed around the injected sites by immunohistochemical examination. CONCLUTION Allogeneic BMMNCs transplantation regulate the expression of Bcl2, Fas and FasL and inhibits cardiomyocyte apoptosis after AMI.
