摘要:
目的 研究牛磺酸在心肌低温冷缺血损伤中的保护作用。方法 SD大鼠随机分成2组,包括对照组(HTK液保存),牛磺酸组(HTK液中加15 mmol/L牛磺酸)。利用离体鼠心非循环式Langendorff 灌注模式装置,经4℃保存4h后,观察复温再灌时的冠脉漏出液中心肌酶的漏出量,并切取心肌组织做光镜和电镜观察。结果 与对照组比较,牛磺酸组能减少心肌酶(乳酸脱氢酶、磷酸肌酸激酶及谷草转氨酶)的漏出,减轻线粒体的损伤。结论 牛磺酸对心肌的冷缺血损伤具有保护作用。
Abstract:
AIM To investigate the effects of taurine on heart hypothermal preservation. METHODS Twenty SD rats were divided randomly into 2 groups: Control group, in which hearts were arrested and stored in HTK solution and Taurine group, in which hearts were arrested and stored in HTK solution containing taurine (15 mmol/L). Langendorff model perfusion apparatus of isolated rat heart was used. The isolated rat hearts were stored in 4℃ in different preservation solutions for 4 hours respectively, and then were reperfused. After 4 minutes' reperfusion, the leakage of myocardial enzyme in coronary effluent was determined. After 10 minutes' reperfusion, the heart samples were obtained and observed under lighter and electronic microscopes. RESULTS Compared with that in control group, the leakage of myocardial enzymes (lactate dehydrogenase, creatine kinase and glutamatoxalacetat transaminase) in the coronary effluent was significantly reduced and the damage of mitochondria was slight in the taurine group. CONCLUSIONT aurine can protect the myocardium in the heart hypothermal preservation.
