摘要:
目的 观察人高脂血清及阿托伐他汀对主动脉平滑肌细胞(smooth muscle cells, SMCs)增殖的影响。方法 以组织块法培养SD大鼠主动脉SMCs,取第5~15代 SMCs 用于实验。按加入干预因素不同分为10组,第1~5组分别加入胎牛血清、人正常血清、高胆固醇血清、高三酰甘油血清及高胆固醇且高三酰甘油血清。第6~10组在1~5组基础上分别加入100 μmol/L阿托伐他汀。采用MTT比色法测定细胞数目。结果①胎牛血清组、正常血清组、高胆固醇血清组、高三酰甘油血清组、高胆固醇并高三酰甘油血清组干预48 h后,MTT A490值分别为0.484±0.034、0.486±0.012、0.535±0.014、0.498±0.030、0.573±0.004,其中高胆固醇血清组与正常血清组相比,明显促进SMCs增殖(P<0.01);高胆固醇且高三酰甘油血清组与前4组相比,均明显促进SMCs增殖(P<0.05);②上述各不同质血清组加入阿托伐他汀干预后,MTT A490值分别为0.383±0.012、0.384±0.018、0.411±0.013、0.384±0.008、0.432±0.006。与各自对应的血清组比较,均明显抑制SMCs增殖(P<0.01)。结论 ①高胆固醇血清及高胆固醇且高三酰甘油血清促进SD大鼠主动脉SMCs增殖;②阿托伐他汀可显著抑制高脂血清对SD大鼠主动脉SMCs增殖作用。
Abstract:
AIM To observe the effects of hyperlipemic serum and/or atorvastatin on proliferation of aorta SMCs in SD rats. METHODS SD rats' aorta SMCs were isolated and cultured by attachmentblock culture. The 5th~15th generation of SMCs were used for experiment. By different serum interventions, we divided the SMCs into 10 groups. The 1st5th groups were intervened respectively by fetal bovine serum, human normal serum, serum of high level of cholesterol, serum of high level of triglyceride, and serum of high levels of cholesterol and triglyceride. 100 μmol/L atorvastatin was added to each of the groups above and thus formed the 6th~10th groups. Cell proliferation was evaluated by MTT colorimetry. RESULTS The result showed that: ①After 48 h culture, MTT A490 values of FBS group, normal serum group, high cholesterol serum group, high triglyceride group, and high cholesterol + high triglyceride group were 0.484± 0.034, 0.486± 0.012, 0.535± 0.014, 0.498± 0.030, and 0.573± 0.004, respectively. Among them, A490 values of high cholesterol serum group was significantly higher than that of normal serum group(P<0.01)and high cholesterol + high triglyceride group was the highest(P<0.05); ② A490 values of the 6th~10th groups were 0.383±0.012, 0.384±0.018, 0.411±0.013, 0.384±0.008, and 0.432±0.006, respectively. Compared with their corresponding serum group, atorvastatin inhibited the proliferation of SMCs(all P<0.01). CONCLUSION ①High cholesterol serum and high cholesterol + high triglyceride serum promote SD rats' aorta SMCs hyperplasia; ②Atorvastatin can notably inhibit hyperlipemic seruminduced proliferation of vascular smooth muscle cells of rats.
