目的 研究骨髓间充质干细胞(BMSC)与乳鼠心肌细胞共移植的细胞替代作用,进而防止左室重构,增强心肌梗死后心功能的恢复。方法 制作大鼠心梗模型的大鼠随机分为对照组,心肌细胞组(移植乳鼠心肌细胞),BMSC移植组和共移植组(移植乳鼠心肌细胞和BMSC)。分别在移植前1 d,移植后4周和12周,用超声心动图测量各组大鼠心功能的情况。干细胞移植12周,测量各组大鼠的质量,左室与质量比值和心梗面积并进行比较。结果 移植后12周,心肌移植组,BMSC移植组和共移植组的左室缩短分数(FS)和左室射血分数(LVEF)有所增加(P<005),而对照组则出现数值下降(P<005)。心功能改善:共移植组>BMSC移植组或心肌细胞移植组>对照组。移植后4周, BMSC移植组和共移植组的FS和LVEF有所增加(P<005),而对照组和心肌移植组则出现数值下降(P<005)。心功能改善:共移植组>MSC移植组,对照组和心肌移植组差异不显著。移植12周后,心梗面积,左室质量,室体比值和心脏大小之间比较皆为:对照组>心肌细胞移植组>BMSC移植组>共移植组>正常组。结论 与MSC和心肌细胞单独移植相对比,共移植能够促进心功能恢复,减轻左室质量,缩小心梗面积,进一步防止心梗后的心室重构。
Abstract:
AIM To observe whether cotransplantation of MSC and neonat rat cardiac cell could replace the loss of cardiac cells so as to prevent myocardial remodeling and improve cardiac function after myocardial infarction. METHODS Rats with myocardial infarction were randomized into 4 groups: Control group, Cardiomyocyte (neonate rat cardiac cell) group, BMSC group and Cotransplatation (cotransplanting BMSC and cardiomyocyte) group. Cardiac function was evaluated by Echocardiography at 1d before transplantation, and 4 wk and 12 wk after transplantation respectively. Body weight, LV/Body weight ratio and myocardial infarction size were calculated at 12 wk after transplantation. RESULTS We found that 12 wk after the injection, highest value increase of FS and LVEF was observed in Cotransplatation group, followed by BMSC group and Cardiomyocyte group(P<005), while value decrease of FS and LVEF was seen in the control group (P<005). 4 wk after the injection, the indexes for the increases of FS and LVEF in Cotransplatation group were higher than those in the BMSC group, which were higher than those in Cardiomyocyte group and Control group (P<005), but no significant difference was seen between Cardiomyocyte group and Control groups. Consistent with the changes in LV weight, Body weight, LV/Body weight ratio and heart infarcted size at 12 wk after injection were significantly lower in BMSC group and Cotransplantation group than those in Control and Cardiomyocyte group, and the maximal decrease was seen in Cotransplantation group. CONCLUSION Compared with transplantation of MSC or neonat rat cardiac cell, cotransplantation improvs cardiac function, attenuates LV weight and decreases myocardial scar size, thus preventing myocardial remodeling after myocardial infarction.
