目的 研究慢性多重应激大鼠脱离应激源后心电图(Electrocardiogram,ECG)变化和下丘脑-垂体-肾上腺(hypothalamicpituitaryadrenocortical,HPA)轴应激反应。方法 健康雄性SD大鼠20只随机分为应激组和对照组。选用刺激脉冲随机变动的噪声、足底电击、强迫游泳、束缚和夜间光照的复合刺激为应激组大鼠应激源,应激刺激持续30 d,建立大鼠慢性多重应激模型。于应激刺激前和完全脱离应激源后3 d记录心电图,分析血清促肾上腺皮质激素(ACTH)和皮质酮(corticosterone,Cor)的变化。结果 刺激4周后,脱离应激源3 d,在1 h心电图记录期中,大鼠心律失常类型和次数明显多于应激前和对照组:持续窦性心动过速或窦性不齐、房性期前收缩(应激前14±07 vs 应激后402±5,P<001)、室性期前收缩(应激前31±06 vs 应激后557±4,P<001)、阵发性室速(应激前0 vs 应激后238±6,P<001)。心率明显增快[(3713±12)次/min→(5087±10)次/min, P<001]、RR间期明显缩短[(1716±5)ms→(1115±05)ms, P<001]、ST段抬高或下移、T波高耸直立或低平,血清ACTH[(531±4)ng/L→(2347±16)ng/L,P<001]和Cor[(16±4)μg/L→(279±50)μg/L,P<001]水平明显增高。结论 慢性多重应激大鼠在脱离应激源后仍存在心律失常,并且HPA轴应激反应增高。
Abstract:
AIM To study the effect of chronic multistress on ECG and HPA axis activity in rats after exposure to multiple stressors. METHODS In this study a chronic multistress animal model was established in rats and the animals were subjected to 4 weeks of chronic multistress. Electrocardiographic recordings were performed before (baseline, B) and after (poststress, PS) chronic multistress. HPA axis activity was examined by collecting blood samples, which were analyzed for plasma ACTH and corticosterone (Cor) concentrations. The effect of chronic multistress on electrocardiogram (ECG) and serum ACTH and Cor was analyzed. RESULTS After 4 weeks of chronic multistress, a much higher incidence of arrhythmias including sinus tachycardia, sinus arrhythmia, atrial premature beats (B:14±07 vs PS: 402±5, P<001), ventricular premature beats(B:31±06 vs PS: 557±4, P<001) and ventricular tachycardia (B: 0 vs PS: 238±6, P<001) was produced. The chronic multistress also induced a much stronger increase of heart rate \[(3713±12)/min→(5087±10)/min, P<001\] and a reduction of average RR interval \[(1716±5) ms→(1115±05) ms, P<001\]. The ST of ECG was lightly displaced, T wave of ECG was upright lofty and serum ACTH \[(531±4)ng/L→(2347±16)ng/L, P<001\], Cor \[(16±4)μg/L→(279±50)μg/L, P<001\] level was raised compared with those in baseline and the control groups. CONCLSUSION Chronic multistress causes arrhythmias and increases HPA axis activity in rats following their exposure to multiple stressors.
