摘要:
目的 探讨补充生理水平的外源性雄激素对去势大鼠动脉壁内皮型NO合酶(eNOS)和诱导型NO合酶(iNOS)表达的影响。方法 将大鼠随机分为去势组、外源性生理水平雄激素补充组和对照组。采用实时荧光定量PCR和Western blot,观察不同处理组胸主动脉壁一氧化氮合酶(NOS)亚型eNOS和iNOS mRNA和其蛋白表达的变化。结果 与对照组相比,去势组大鼠eNOS的表达显著降低(P<0.05),而外源性雄激素生理水平补充组大鼠eNOS的表达接近正常水平;两个处理组iNOS mRNA的水平与对照组比较均无统计学差异。结论 补充外源性生理水平的雄激素对去势大鼠心血管系统的保护作用,可能是通过血管壁eNOS影响NO的合成与释放而介导的。
Abstract:
AIM To investigate the effects of exogenous androgen at physiological level on the expression of eNOS and iNOS in the aortic wall of orchiectomized rats. METHODS Animals were randomly divided into 3 groups: orchiectomized group, androgen treated at physiological level group and the sham control. Real-time RTPCR and Western Blotting were used to detect the expression of eNOS and iNOS at the mRNA and protein levels, respectively. RESULTS Compared with that in sham control group, the expression of eNOS in the orchiectomized group was significantly downregulated (P<005), but the expression of eNOS in the androgen treated group did not decrease. No significant difference in iNOS expression was found between the groups. CONCLUSION The protective effect of exogenous androgen at physiological level in orchiectomized rats is probably mediated by the synthesis and release of NO affected by eNOS of blood vessel endothelium.
