摘要:
目的 应用原子力显微镜(AFM)观察动脉粥样硬化(AS)时血管内皮细胞(VEC)膜超微结构的变化。方法 取新西兰纯种雄性大白兔24只,随机分为2组,正常对照组12只(给予普通饲料)、高脂模型组12只(给予高脂饲料),于6周末处死。取胸主动脉中段制作标本,应用AFM进行扫描。结果 50 μm范围扫描对照组VEC呈梭形,排列规整,其长轴与血液流动方向一致。高脂模型组细胞明显肿胀变形,形态呈球形、不规则形、体积变大,排列紊乱。500 nm扫描范围对照组细胞膜由圆形、椭圆形等大小较一致的颗粒样隆起构成;隆起结构光滑、饱满,排列疏密均匀一致,各隆起间界限分明。高脂模型组细胞膜由长条形、不规则形大小相差悬殊的隆起构成;隆起表面凹凸不平,排列疏密明显不均;各隆起间界限变得模糊不清。结论 利用AFM可清晰地显示AS时受损的VEC膜的超微结构变化。
Abstract:
AIM To observe the ultrastructure of endothelial cell membrane in atherosclerotic rabbits by atomic force microscope (AFM). METHODS A total of 24 male New Zealand white rabbits were randomly divided into 2 groups: control group of 12 rabbits and AS group of 12 rabbits. By the end of 6th week the rabbits were sacrificed and the middle segments of thoracic aortas were obtained and observed under atomic force microscope. RESULTS The vascular endothelial cells (VECs) in the control group were fusiform in shape and aligned regularly. Their macroaxis were paralleled with the direction of hemokinesis. VECs in the atherosclerotic group were in deformity and bigger than those in the control group. They aligned irregularly and their volumes became swollen. The membrane protein of VECs in the control group was composed of many round and elliptical eminences. The eminences were almost of the same size and were distinct among the eminences. The membrane protein of VECs in the atherosclerosis group was composed of many irregular eminences. The size of the eminences was different. They were vague among the eminences and had many holes. CONCLUSION The changes of the ultrastructure of the membrane surface in the damaged VECs in AS are directly observed by AFM.
