摘要:
目的: 探讨自制的新型磁性微球聚乳酸复合材料及载药雷帕霉素磁性微球聚乳酸复合材料对血管内皮细胞增殖的影响。方法: 将不同含量的磁性聚合物微球聚乳酸复合材料和磁性聚合物微球的载药雷帕霉素聚乳酸磁性复合材料放置于培养的血管内皮细胞溶液中,标准条件下共培养4 d后,四噻唑蓝比色法测定细胞数量,倒置显微镜下观察内皮细胞的增殖情况。结果: 培养1、2、3、4 d的磁性微球聚乳酸复合材料不同浓度组对血管内皮细胞生长均有明显的促进作用(P<0.05)。雷帕霉素是抑制内皮细胞生长,但是载药的磁性微球聚乳酸复合材料各浓度组内皮细胞的数量显著多于单纯药物对照组(P<0.05),磁性复合材料显著的减弱了雷帕霉素对内皮细胞的抑制作用。结论: 磁性微球复合材料对血管内皮细胞具有促进生长的作用。
Abstract:
AIM: To investigate the effects of new magnetic composite materials regarding magnetic microsphere (MMS) and poly(L-lactide) (PLLA) on proliferation of vascular endothelial cells (VEC). METHODS: Different contents of MMS/PLLA and rapamycin/MMS/PLLA were added to the cell culture medium and kept under standard conditions for 4 days. MTT was used to determine the amount of VEC proliferation. RESULTS: The amount of the VEC proliferation was significantly higher than in the control group (P<0.05). There was a higher proliferation of rapamycin/MMS/PLLA than rapamycin/PLLA (P<0.05). Magnetic composite materials significantly weakened the inhibitory effect of rapamycin on endothelial cells. CONCLUSION: Magnetic composites may promote the growth of vascular endothelial cells.
