摘要:
目的 通过观察不同浓度三氧化二砷(As2O3)对内皮细胞形态及增殖活力的影响,从内皮细胞角度评价As2O3防治经皮冠状动脉成形术(PTCA)后再狭窄的作用。方法 以不同浓度As2O3(0.01 μmol/L、0.1 μmol/L、1 μmol/L和10 μmol/L)作用于传代培养的人静脉内皮细胞株(EVC-304),分别培养24 h、48 h和72 h观察细胞形态、绘制细胞生长曲线,并行MTT比色法观察As2O3对内皮细胞增殖活力〔吸光度(A)值〕的影响。结果 10 μmol/L的As2O3使细胞皱缩变圆,甚至浮起、碎裂。≤1 μmol/L的As2O3作用于内皮细胞后,细胞形态无明显变化。细胞计数显示,As2O3浓度高于1 μmol/L时,内皮细胞数明显降低。MTT比色法显示:As2O3浓度低于1 μmol/L,A值与对照组比较无差异,在1~10 μmol/L浓度范围内,A值明显降低(P<0.05),提示细胞增殖活力受到抑制。结论 高浓度As2O3对内皮细胞形态有破坏作用,并显著抑制内皮细胞增殖,且抑制作用具有时间及剂量依赖性。低浓度的As2O3则对内皮细胞形态及增殖活力无影响。
Abstract:
AIM To investigate the effect of arsenic trioxide (As2O3) on cellular morphology and proliferation of endothelial cells and to evaluate its value in inhibiting restenosis. METHODS Human umbilical endothelial cells (ECV-304) were incubated with As2O3 in vitro (0 μmol/L, 0.01 μmol/L, 0.1 μmol/L, 1 μmol/L and 10 μmol/L). Cell morphology was observed by inverted microscopy, cell survival number was described by cell growth curve and cell viability was measured by MTT method. RESULTS After incubation, paste wall cells decrease and float and vacuolation were observed in endothelial cells in 10 μmol/L As2O3 group. As2O3 of 0.01-0.1 μmol/L exerted no effect on the A value of endothelial cells. However, the A values of endothelial cells incubated with As2O3 of 1-10 μmol/L were lower than those in control group, indicating that As2O3 inhibited the proliferation of endothelial cells. CONCLUSION As2O3 (10 μmol/L) damages cellular morphology and inhibits cell proliferation.
