摘要:
目的 评价糖尿病对大鼠心室肌细胞肌浆网钙离子转运通道及其调控蛋白表达的影响。方法 以注射链脲佐菌素的方法制备1型糖尿病大鼠模型;动物分成4组(n=6):对照组、糖尿病4周组、糖尿病8周组和糖尿病12周组。分别以RT-PCR和Western blot的方法测定心室肌细胞肌浆网上主要钙转运通道及其调控蛋白的mRNA和蛋白的表达变化。结果 糖尿病程第4周,心肌细胞肌浆网上主要钙转运通道(雷诺定受体和Serca2ATPase)表达开始显著下降,并持续下降至病程12周。同时,调控蛋白FKBP12.6表达第8周显著下降、phospholamban表达同步上升。结论 随着病程的进展,糖尿病大鼠心肌细胞肌浆网钙离子转运通道及其调控蛋白的表达呈逐步和显著的下降趋势。
Abstract:
AIM To evaluate the effects of diabetes on the expressions of Ca2+ channels and regulatory proteins on sarcoplasmic reticulum (SR) in rat cardiomyocytes. METHODS Sprague Dawley rats were divided into four groups (n=6, each group): group A (control group) and streptozotocin-induced diabetic groups: group B (4-week group), group C (8-week group), and group D (12-week group). Expressions of Ca2+ channels and their regulatory proteins were assessed by RT-PCR (mRNA) and Western blotting (proteins), respectively. RESULTS With the progression of diabetes, expressions of both ryanodine receptor 2 (RyR2) and sarcoplasmic reticulum Ca2+-2ATPase (Serca) decreased significantly, with the downregulation of FKBP12.6 and upregulation of phospholamban. CONCLUSION With progression of disease, diabetes induces the declines in expressions of Ca2+-associated channels and regulatory proteins on SR in cardiomyocytes of rats, gradually and significantly.
