作者利用 Balb/ c小鼠的血管紧张素 的 1型受体(AT1 )基因和兔 AT1 基因的序列的不同 ,合成了一个新的含有两种动物序列的内标准 RNA,有效地消除了二级结构对反转录 PCR(RT- PCR)的影响。本研究对利用这种内标准 RNA进行定量 RT- PCR进行初步评价。
Abstract:
In this test,we synthesized an internal standard RNA,which have part sequences of receptor type 1 of ATⅡ (AT 1) gene of the Balb/c mouse and rabbits,utilizing difference of the AT 1 gene sequence in these two animals,This internal standard RNA abolishes the influence of the secondary struture in the reverse transcription polymerase chain reaction (RT PCR). In the standard curve estimation by the quantitative RT PR using this internal standard RNA,the correlation coefficient is 0.998. From above data,it is suggested that the AT1 expression detecting by quantitative RT-PCR using this internal standard RNA can be truly reflected and the quantitative RT-PCR using this internal standard RNA is a credible method. Moreover, this internal standard RNA can be made simply and speedily.
