摘要:
目的 观察tirofihan对犬髂动脉球囊损伤后血浆可溶性牯附分子CD62p水平及全血白细胞计效的影响。方法 21只健康成年犬随机分为治疗组(tlrofihan 300~g／kg静推．后以0．3 pg·kg ·rain叫静滴6 h)和对照组(9g／L盐水静漓)，每组l2只．行单侧髂动脉球囊损伤，术前、术后即刻及2。4，6 h静脉取血行全血白细胞计效。ELISA法测定血浆CD62p含量(光密度)。结果 对照组球囊成形术后血浆CD82p古量增加，2 h后开始下降}而治疗组CD6Zp术后立即下降。4 h后恢复至基线。全血白细胞在对照组术后逐渐增加i而治疗组用药后则出现一过性白细瞧受抑。结论 tirofiban除了已知的抑制GPI b／I a受体和血小板聚集外，还能抑制球囊成形术后血小板活化及全血白细胞．可能通过抑制CD62p(P—seleetin)、抑制白细胞一血小板问相互作用改善经皮冠状动脉舟入治疗后的临床结果．
Abstract:
AIM To observe the effects of GP I b／I a antagonist tirofoban on level of plasma soluhie adhestion molecule CD62p and whole blood leukocyte counts in canine model of iliac artery injury by balloon angioplasly．METHODS 24 health adult dogs were randomized into the treated group(tirofiban 300#g／kg bolus+O．3 ·kg ·min continuous venous injection／or 6 h)and the control group(treated with saline)．1liac artery injury by balloon angioplasty was performed 15 min after the drug injection．Blood samples were drawn through femoral vein catheter．Whole blood 1eukocyte counts were measured mechanically and plasma CD62p concentration(A)was measured by El，ISA 15 min before and after procedure every 2 h／or 6 h．RESULTS CD62p leve1 was increased after angioplasty and began to decrease 2 h after the procedure in the control group．while reduced immediately and then increased to baseline 4 h after angioplasty in the treated group．W hole blood leukocyte counts were increased after the procedure in the control group，whereas a transient decrease after the administration of tirofiban was observed in the treated group．CONCLUSION In addition tO its known effect on platelet aggregation by block GPⅡh／Ia receptor，tirofiban can inhibit platelet activation after angioplasty and decrease leukocyte counts transiently．The outcome improvement On percutaneous coronary intervention by tiroflban may be acquired through inhibition of CD62p(P—selectin)and interaction of leukocyte and platelet．
