摘要:
目的 探讨高脂饮食兔血管内皮功能的早期变化。方法 20 只高脂饮食兔分别于实验前及实验第1, 2, 3, 4 周随机抽取4 只, 颈内静脉插管, 抽血测血脂、基础一氧化氮(NO ) 水平, 随后颈内静脉内注射1 ml.kg- 1乙酰胆碱(Ach, 100 nmol.L - 1) , 5 m in 后从插管处抽血测Ach 刺激后NO 水平。结果 随着胆固醇的逐渐上升, 基础NO 水平与Ach 刺激后NO 水平均下降, 与实验前比较, 基础NO 水平至实验2 周时才出现差异(P < 0. 05) , 而Ach 刺激后NO 水平在实验1 周时即出现差异(P < 0. 05)。结论 胆固醇水平升高导致血管内皮功能受损, 内皮功能障碍首先表现为内皮储备功能障碍, 接着出现静息时内皮细胞障碍。
Abstract:
AIM To study the early change of vascular endothelial function in hyperlipid diet rabbits . METHODS Four rabbits were randomly selected from 20 rabbits before experiment and 1 week, 2 week, 4 week and 8 week after treatment with high lipid diet. A catheter was inserted into the right jugular vein and the blood samples were drawn inorder to measure serum lopids and NO. NO was also examined 5 minutes after 1 m l.kg- 1 acetylcholine (Ach ) (100 nmol.L- 1) was in travenously injected. RESULTS The NO production was gradually reduced in rabbits treated with hyperchlesterol diet as time passed. Compared with those before experiment, basal NO production had no difference in the first week, and shown to bestatistical difference in the second week (P < 0. 05) ; However, NO production stimulated by Ach was shown to be different in the first week (P < 0. 05). CONCLUSION Hypercholesterolemia results in the endothelial dysfunction. This endothelial disorder is characterized by the impairment in Ach stimulated NO formation firstly with subsequent injured basal NO production, a phenomenon with the early impaired endothelial preserved function.
