摘要:
目的 观察原代培养的乳鼠心肌细胞的形态学特征。方法 使用II型胶原酶、胰蛋白酶和DNaseI 消化心脏组织, 差速贴壁分离法和化学试剂抑制法纯化心肌细胞。结果 第1 天心肌细胞几乎全部贴壁, 少数细胞出现自发性搏动; 第3 天形成细胞簇或细胞单层, 收缩明显而有力; 透射电镜可见肌小节、Z 线、闰盘; 良好的形态和活力可维持 20 d。结论 使用II 型胶原酶、胰蛋白酶和DN ase? 分离培养心肌细胞可获得形态、活力良好的心肌细胞, 该方法值得进一步推广。
Abstract:
AIM To observe the morphological characteristics of the cultured cardiom yocytes.METHODS The hearts were digested by collagenase type II , trypsin and DN ase I. The cell suspension was preplated to reduce the fibroblast content. The cells were treated with bromodeoxyuridine (Brdu) for 3 days. RESULTS The cells spread over the first day and developed their characteristic morphology by day 3 or 4. They were arranged in loose and interlacing networks of single cells. Cross-striations, Zband and in tercalated disc were observed with transmission electron microscopy. The cells beat spontaneously at 40～ 110 perminute and continued 20 days.CONCLUSION It can obtain cardiomyocytes with well morphology and spontaneous beating with collagenase type II , trypsin and DN ase I. This method is worth of spreading.
