摘要:
目的:探讨低压缺氧时与内皮细胞相关的一氧化氮(NO),内皮素(ET-1)、肾上腺髓质素(ADM)的变化,了解低压缺氧的损伤机制。方法: 将30只雄性Wistar大鼠, 随机分为5组:对照组、低压缺氧后30 min, 6 h组、使用N-甲基-L-精氨酸(L-NAME)保护后30min和6h组, 每组6只。实验组大鼠在低压舱内模拟上升至8000m、停留30min, 重复1次。分别于缺氧后30min和6h 处死大鼠取血及心脏、肺组织, 测定NO,ET-1,ADM及半定量测定心脏及肺组织eNOS mRNA,iNO SmRNA。结果:低压缺氧后大鼠血中NO,ET-1及心脏和肺组织iNOS mRNA 显著增高,ADM, eNOS mRNA显著降低。注射L-NAME后可明显减少NO ,ET-1,ADM的变化值。结论:NO在低压缺氧后对ADM, ET-1的变化有影响。
Abstract:
AIM: To study changes of NO, ET-1, ADM and mRNA expression of eNOS, iNOS after acute hypoxia. MEHHODS: Thirty conscious Wistar rats were randomly divided into 5 groups: 1 control group and 4 experim ental groups. Two of experimental groups were injected N-M ethyl-L -Arginine (L-NAME, 100mg?kg) before experiment. The experimental groups underwent 8000m altitude exposure in hypobaric chamber for 30min, and repeat it after 0m altitude. The rat blood and heart and lung were taken 30min, 6h after exposure and NO, ET-1, ADM and eNOS mRNa, iNOS mRNA expression level in rat heart and lung were measured. RESULTS: The level of NO ,ET-1 in the experimental groups were significantly higher and ADM was significantly lower than that of control group, and these changes can be rest rain in injected L-NAME experimental groups. CONCLUSION: NO may play an important role in the change of ADM , ET-1 after acute hypoxia.
