摘要:
为探讨压力负荷增高性心肌肥厚大鼠心肌胶原网络的重构及其与左室功能的关系。采用腹主动脉结扎术致大鼠心肌肥厚; 心脏离体灌注测定左室功能; VG染色和图像处理观察心肌胶原网络重构。结果显示手术组大鼠术后2周即出现明显左室肥厚; 与假手术组比较, 左室心肌总胶原容积百分比(CVF-T)于术后2周即呈增高趋势(P<0.05) 术后4.8周CVF-T和无小血管视野CVF(CVF-NV) 均显著高于假手术组; 手术组大鼠术后4周左室功能出现明显减退, CVF-NV,CVF-T与左室功能指标, 尤其舒张功能指标呈明显负相关。结论: 压力负荷增高性心肌肥厚时伴有心肌胶原网络的重构, 心肌胶原网络重构与心功能障碍, 尤其是舒张功能障碍有明显关系。
Abstract:
To examine myocardial collagen network remodelling and its relation with changes of left ventricular function in pressure-overloaded hypertrophy rats, the abdominal aorta banding was used to induce hypertrophy in rats. Left ventricular function was monitored with Langendorff preparation. Myocardial collagen was stained with VG and analyzed using a computer assisted procedu re. The results showed that the operated rats manifested myocardial hypertrophy only 2 weeks af ter the operation . In comparison with sham-operated rats,myocardial total-Collagen Volume Fraction (CVF-T) increased 2 weeks after the operation (P < 0.05) , CVF-T andnonvascular-Collagen Volume Fraction (CVF-NV) both increased significantly 4 and 8 week safter the operation (P <0.01). In the operated rats, left ventricular function was reduced significantly 4 weeks after the operation. CVF-NV , CVF-T was negatively correlated with the parameters of left ventricular function,especially those of diastolic function. It may be concluded that there occurs a myocardial collagen network remodelling in ressureoverloaded hypertrophy and that myocardial collagen network remodelling is probably related to ventricular dysfunction, especially diastolic dysfunction.
