用膜片钳全细胞记录法研究关附甲素(GA) 对分离的单个豚鼠心室肌细胞钠通道电流(INa) 和L-型钙通道电流(ICa-L) 的影响。结果表明: 8.40 umol/LGA 使INa峰值( INamax) 从8.02±2.53 nA 分别降至5.79±1.82 nA 和4.44±1.81 nA (n=8, P < 0.05 和P <0.01) ; 使ICa-L最大峰值( ICa-Lmax ) 从1020.8±197.3 pA 分别降至523.0±101.8pA和429.6±120.0 pA (n=5, 均P <0.01)。GA 使INa和ICa-L的电流-电压曲线上移, 但均不改变其激活、峰值和反转电位。本研究表明: GA 对INa和ICa-L具有浓度依赖性阻滞作用, 这种阻滞作用可能为其抗心律失常作用的主要机制。
Abstract:
The effects of Guanfu base A (GA) on sodium channel current INa) and L-type calcium channel current (ICa-L) were investigated in isolated single ventricular cells from guinea pig hearts by means of patch clamp whole-cell recording. The results showed that GA at 8, 40 umol/L decreased peak INa(Iinf>Namax) from 8.02±2.53 nA to 5.79±1.82 nA and 4.44±1.81 nA (n=8, P <0.05 and P <0.01) , reduced peak ICa-L( ICa-Lmax ) from 1020.8±197.3 pA to 523.0±101.8 pA and 429.6±120.0 pA (n= 5, P < 0.01). GA shift up boh the I-V curve of INa and that of ICa-L, but didn′t change their active, peak and reverse potentials. It reveals that GA blocks both INa and ICa-Lin a concentration dependent manner, which might be the major mechanism of its antiarrhythmic action.
