摘要:
目的 观察血小板衍化生长因子(PDGF)对培养的血管平滑肌细胞(VSMC)增殖及胶原蛋白合成的影响。方法 采用培养的免动脉VSMC,应用 H-TdR的 H-脯氨酸掺入方法,观察PDGF-BB对免VSMC DNA合成以及胶原蛋白合成的影响。结果 PDGF-BB可促进处于静止状态的兔VSMC DNA及胶原蛋白的合成,并呈现明显的浓度依赖关系,在40 ug/L的浓度时DNA及胶原蛋白的合成达到高峰,DNA及胶原蛋白分别处于36 h和48 h合成最为显著。结论 PDGF-BB可明显促进培养的VSMC增殖及胶原蛋白的合成。
Abstract:
AIM To investigate the effects of platelet-derived growth factor(PDGF)on proliferadon and coilagen protein synthesis of vascular smooth muscle cells(VSMC).METHODS In the present experiment,the rabbit VSMC were cultured and effects of PDGF-BB on DNA synthesis and collagen protein synthesis of rabbit VSMC were observed by using H-TdR incorporation and H-proline intorporation in vitro.RESULTS PDGF-BB could significantly promote DNA synthesis and collagen protein synthesis of quiescent rabbit VSMC in a dose-dependent manner,with a maximal response at a concentration of 40 ug/L at 36 h and 48 h respectively.CONCLUSION PDGF-BB could promote proliferation and collagen protein synthesis of cultured rabbit VSMC.
