摘要:
目的 研究牛磺酸对培养的心肌细胞模拟缺血/再灌注( I/R ) 过程中心肌细胞凋亡的影响。方法 培养SD大鼠乳鼠心肌细胞, 建立模拟/R 模型。实验分3 组: ①正常对照组; ②模拟I/R 组: 缺氧120min, 复氧240min; ③牛磺酸组: I/R 前加用20mmol/L牛磺酸。计算台盼蓝摄取率, 测定乳酸脱氢酶(LDH) 活性, 以流式细胞仪(FCM) 和透射电镜观察细胞凋亡。结果 在正常对照组, FCM 未发现凋亡峰, 电镜未发现凋亡细胞; I/R 组FCM 检测的凋亡率为15.1% , 电镜下可见到典型的凋亡细胞, 台盼蓝摄取率和LDH 活性较对照组显著增加[分别为(35.3±1.7)%vs(2.6±0.8)% 和(184.8±4.6) U/ml vs (46. 3±2. 4) U/ml; 两者P<0.05 ]。牛磺酸组心肌细胞大部分存活, 凋亡率明显降低(平均3.5%,P<0.05)。结论 细胞凋亡参与了心肌I/R 损伤, 牛磺酸可减少I/R 心肌细胞的凋亡。
Abstract:
AIM To examine the effects of taurine on apoptosis in cultured neonatal rat cardiomyocytes induced by simulated ischemia-reperfusion. METHODS A cell culture model of neonatal rat cardiac myocytes was used. The cultured cardiomyocytes were classified into three groups, ie, control group, simulated ischemia-reperfusion group(anoxia for 120 min,reoxygenation for 240 min) and taurine group(20 mmol/L taurine was added before anoxia). The trypan blue absorbance and LDH activity were detected.Apoptosis was determined by flow cytometry (FCM )and transmission electron microscopy (TEM ). RESULTS Apoptosis was not found in the control group. After 120 min simulated ischemia followed by 240min reperfusion, typical apoptotic cells were seen by TEM , and FCM showed an apoptosis rate of 15. 1%. While intaurine group , apoptosis rate was decreased to only (3. 5±1. 2)%. CONCLUSION Myocardial apoptosis is involved in ischemia reperfusion injury, and taurine can inhibit cardiomyocyte apoptosis induced by ischemia reperfusion.
