摘要:
目的 观察急性容量超负荷心肌肥大(MH) 早期局部心肌碱性成纤维细胞生长因子(bFGF) 及血管紧张素Ⅱ(AngⅡ ) 的作用以及AngⅡ1 型受体(AT1) 拮抗剂(Lo sartan,Los) 的逆转MH 作用。方法 选择36 只雄性SD 大鼠随机分为3 组: 腹腔动静脉造瘘容量超负荷组(AVO 组)、容量超负荷治疗组(LO S 组) 及对照组(CON 组) , 每组12只, 对比检测术后第10 天和第20 天的收缩压(SBP)、左室重量指数(LVMI)、心肌羟脯氨酸(HY)、bFGF,AngⅡ 及血浆AngⅡ 含量。结果 ①ACF 组SBP 和LVMI 第10 天和第20 天持续升高(P < 0. 05, vsCON 组) , 给药后二者水平仍显著高于对照组, 但较ACF 组明显下调; ②ACF 组心肌bFGF 和A ng? 第10 和第20 天明显上升(P <0105, vs CON 组) ,A T 1 受体被阻断后第10 天心肌bFGF 显著升高(P < 0. 05, vsACF 组) , 第20 天无明显变化(P> 0. 05, vsACF 组) , 而心肌和血浆AngⅡ 较ACF 组明显增多(P < 0. 05) ; ③心肌bFGF,AngⅡ 及LVMI 之间均呈显著正相关。结论 心肌bFGF 可能和AngⅡ 共同参与急性容量超负荷MH 早期发生, 其产生可能与心肌A ng? 有关,A T1 受体拮抗剂早期具显著逆转MH 作用。
Abstract:
AIM To determine the effects of myocardial basic fibroblast growth factor(bFGF), angiotensinⅡ(AngⅡ) and AngⅡreceptor 1(AT1) antagonist Losartan(LOS) on myocardial hypertrophy(MH) in the early stage of acute volume overload(AVO).METHODS Abdominal aorta-inferior vena cava fistula was performed in the rats of group AVO(n=12) and group LOS (n=12,Losartan was given once daily). All the indexes were measured at the 10th and 20th day and were compared with group Control(CON,n=12).RESULTS ①The levels of systolic blood pressure and left ventricle mass index(LVMI) were elevated evidently in group AVO and group LOS compared with group CON (P< 0. 05), while the levels in group LOS were lower significantly than in group AVO;②myocardial bFGF and AngⅡincreased obviously in group AVO (P< 0.05,vsgroup CON). Compared with group AVO,myocardial bFGF increased apparently at the 10th day(P<0.05),but did not change at the 20th day in group LOS(P>0.05),however,myocardial and plasma AngⅡof group LOS were all elevated distinctly(P<0.05, vsgroup AVO);③There was obviously positive correlation among myocardial bFGF,AngⅡand LVMI. CONCLUSION Myocardial bFGF and AngⅡmay play an important role in early course of MH,and AT1 receptor antagonist (Losartan) may reverse MH.
