摘要:
目的:研究人动脉粥样硬化病变部位B族I型清道夫受体(SR-BI)表达增加的机制。方法:采用地高辛标记的寡核苷酸探针原位杂交技术,观察氧化型低密度脂蛋白(OxLDL)及A族清道夫受体(SR-A)的特异度抑制性配体-- 岩藻多糖作用下,原代培养的人外周血单核细胞SR-BImRNA表达与单核细胞趋化蛋白-1(MCP-1)、碱性成纤维细胞生长因子(bFGF)、血小板源性生长因子(PDGF)、白细胞介素-10(IL-10)四种致炎细胞因子mRNA表达之间的关系。果:OxLDL(10~30μg·ml-1)同人外周血单核细胞孵育24h后能够以剂量依赖性方式显著提高SR-BImRNA,MCP-1,bFGF,PDGF与IL-10四种致炎细胞因子mRNA的表达(均P<0.01)。岩藻多糖(50~250μg·ml-1)能够明显抑制OxLDL(20μg·ml-1)对SR-BImRNA及上述四种因子mRNA表达的诱导(均P<0.01)。结论:OxLDL可以同步刺激原代培养的人血单核细胞表达SR-BImRNA和致炎细胞因子mRNA,SR-A是介导此过程最关键的受体。
Abstract:
AIM: To investigate the mechanism of scavenger receptor class Btype I (SR-BI) upregulated in human atherosclerotic lesions. METHODS: With the digoxenin-labeled oligonucleotide-probes in situ hybridization, we investigated the relation between the primary cultured human peripheral blood monocyte expression of SR-BImRNA and expression of proinflammatory cytokines mRNA including MCP-1, bFGF, PDGF and IL-10 affected by oxidized low density lipoprotein (OxLDL) and fucoidin, a specific inhibitory ligand for scavenger receptor class A (SR-A). RESULTS: Monocyte was significantly increased the mRNA expression of SR-BI, MCP-1, bFGF, PDGF and IL-10 in a dose-dependent manner after incubating with OxLDL (10~30μg·ml-1) for 24h (P<0.01). Fucoidin (50~250μg·ml-1) distinctly inhibited OxLDL (20μg· ml-1) from inducing monocyte them RNA expression of SR-BI as well as expression of above proinflammatory cytokines (P< 0.01). CONCLUSION: OxLDL can simultaneously stimulated the primary cultured human peripheral blood monocyte expression of SR-BImRNA and mRNA expression of proinflammatory cytokines, among which SR-A plays a crucialrole.
