目的:评价环氧化酶抑制剂对心肌缺血损伤(AIM)的保护作用。方法:将成年家兔(1.8~2.5kg)24只随机分为正常对照组(A组),单纯心肌缺血组(B组),心肌缺血加迪克乐克组(C组),建立AIM 模型,给予相应的干预,术后6h观察血液学指标,缺血心肌的HE染色及COX-2的多克隆抗体免疫组织化学染色进行病理观察。结果:缺血心肌中COX-2高度表达,血液学指标示术后6h白细胞计数在B、C组间有明显差异(P<0.05),iNOS测定值在A、B组有显著差异(P<0.05), MDA、SOD含量在A、B组间有显著性差异(P<0.05),C组应用非选择性环氧化酶抑制剂,在术后6h白细胞计数升高程度与A、B组均有显著性差异(P<0.05),iNOS测定值与B组无显著差异(P>0.05),MDA、SOD含量与B组差异显著(P<0.05)。结论:缺血心肌存在显著的炎症活动,环氧化酶抑制剂对实验性心肌缺血损伤有保护作用。
Abstract:
AIM: To evaluate the effects ofcyclooxygenase inhibitor on acute ischemic myocardium(AIM)inrabbits. METHODS: 24 rabbits were randomly divided into 3 groups: group A: normal controls; group B: myocardial ischemia; group C: myocardial ischemia+diclofenac sodium sustained release tablets. The left circum flexarteries of rabbits were ligated to obtain the animal models of AIM for 6 hours in groups B and C. 6 hours after the ligation, we measured the counts of leukocyte of periphery blood and serum MDA, SOD and iNOS; HE and monoclonal antibody immunohistochemistry staining, of ischemic tissues were investigated. RESULTS: The expression of cox-2 in ischemic myocardium increased. 6 hours after the ligation, the difference in the increased count of leukocyte between groups B and C was significant (P<0.05); iNOS was similar in groups B and C (P>0.05). The differences in MDA and SOD were between groups B and C significant (P<0.05). CONCLUSION: Cyclooxygenase inhibitor can protect the ischemic myocardium.
