摘要:
目的:观察不同程度低氧对家兔心脏成纤维细胞(CFs)增殖与表型的影响。方法:分离、纯化、培养家兔Us,随机分为3组:对照组、中度低氧组和重度低氧组,以四哇盐(MTT)比色法检测细胞的增殖情况,采用激光共聚焦显微镜检测各组细胞内肌动蛋白α-actin的表达。结果:中度低氧组MTT的490nm吸光值(A490 )为0.46±0.05较对照组(0.37±0.04)显著增加(P<0.05 ,n=8),而重度低氧组A490,值为0.31±0.03较对照组显著降低(P<0.05 ,n=8)。激光共聚焦显微镜结果显示,对照组CFs内无明显的。α-actin表达,荧光值为1127±120。中度低氧组细胞内。α-actin表达显著增加,荧光值为1672±193,与对照组比差异显著(P<0.05,n=4)。重度低氧组细胞内α-actin表达较少,荧光值为1355±231,与对照组比无显著差异(P>0.05,n=4),结论:中度低氧促进家兔Us增殖,并且出现肌成纤维细胞的表型特点,而重度低氧具有细胞毒性。
Abstract:
AIM:To investigate the effect of hypoxia on the proliferation and phenotype of cardiac fibroblasts(CFs) from rabbits. METHODS:The purified cardiac fibroblasts were cultured and divided randomly into three groups:control group,moderate hypoxia (MH) group and severe bypoxia (SH) group. 72 h later,MTT method was used to investigate the proliferation of CFs,and α-actin in cardiac fibroblasts was measured by means of laser scanning confocal microscopy.
