目的: 探讨AT1R 反义寡核苷酸(AT1R-AS-ODNs) 对正常大鼠心肌细胞生长的影响。方法: 脂质体介导AT1R-AS-ODNs转染体外培养的SD 乳鼠心肌细胞,Western印迹及原位杂交检测细胞内AT1R 蛋白及mRNA 表达; 流式细胞仪检测细胞内DNA合成; 免疫沉淀法检测c-Jun氨基末端活化蛋白激酶(JNKs)活性; 免疫细胞化学检测核转录因子c-Jun蛋白表达水平。结果: 在脂质体介导下,AT1R-AS-ODNs成功转染心肌细胞; 转染24 h 后,AT1R蛋白及mRNA表达水平分别被下调6017%、5112% (P <0.05) ; 但AT1R-AS-ODNs 转染对细胞内JNKs活性、c-Jun 蛋白表达、DNA 合成无显著影响。结论:AT1R-AS-ODNs 能有效抑制正常大鼠心肌细胞AT1R 表达, 但不改变细胞内AT1R 偶联的信号转导过程, 也不影响细胞的正常生长。
Abstract:
AIM: To explore the effects of angiotensin Ⅱ receptor 1 antisense oligonucleotides(AT1RAS-ODNs)on physiological growth in cardiac myocytes. METHODS: AT1R-AS-ODNs was transfected into cultured neoatal cardiac myocytes mediated by loposome reagent. The expressions of AT1R protein and mRNA were detected by western-blot and in situ hybridization method, respectively. DNA synthesis was determined by fluorescence activated cell sorting. The activity of c-Jun NH2-terminal kinases (JNKs) was evaluated by immuno-precipitation. The protein expression of c-Jun was detected by immunocytochemistry. RESULTS: Encapsuled by liposome, AT1R-AS-ODNs transfection resulted in the significant decrease of AT1R protein and mRNA by 60.7% and 51.2% , respectively. However, antisense had no significant effects on cardiac myocytes’ grwoth including DNA synthesis, JNKs activation, and c-Jun protein expression. CONCLUSION: Despite evident inhibitory effect on AT1R gene expression, AT1R-AS-ODNs had no significant effects on cardiac myocytes’ growth including DNA synthesis and AT1R assoiated signaling transduction in cardiac myocytes.
