摘要:
目的:观察白细胞介素26 (interleukin26 , IL26) 对血管升压素(argipressin , AVP) 诱导的心脏成纤维细胞(CFs) 增 殖及p27 蛋白表达的影响。方法:以培养的新生Sprague2Dawley(SD) 大鼠CFs 为实验模型,四氮唑盐(MTT) 比色法检 测细胞增殖,流式细胞分析仪(FCM) 技术测定细胞周期及p27 蛋白表达阳性率。结果: ①10 000 U/ ml IL26 作用于 CFs 48 h ,MTT法A 值(0. 38 ±0. 01) 显著高于基础状态组(0. 32 ±0. 01 , P < 0. 01) ;10 - 7 mol/ L AVP 和10 000 U/ ml IL2 6 共同作用组CFs 的A 值(0. 41 ±0. 01) 也明显高于AVP 单独作用组(0. 39 ±0. 01 , P < 0. 01) 。②IL26 作用组CFs 的 S 期细胞百分率(13. 0 %±3. 5 %) 和细胞增殖指数(PI) (29. 4 %±1. 9 %) 均高于基础状态组(7. 5 % ±1. 0 %和26. 0 % ±1. 0 % , P < 0. 01) ,G0/ G1 期细胞百分率(70. 6 %±1. 9 %) 低于基础状态组(74. 0 %±1. 0 %) ;AVP + IL26 组CFs 的S 期细胞百分率和PI 分别为18. 8 %±1. 5 %和35. 2 %±1. 6 % ,明显高于AVP 组(15. 5 %±1. 4 %和31. 4 %±1. 5 % , P < 0. 01) ,而G0/ G1 期细胞百分率(64. 8 %±1. 6 %) 明显低于AVP 组(68. 6 %±1. 5 % , P < 0. 01) 。(3) IL26 组CFs p27 蛋白表达阳性率(72. 6 %±2. 5 %) 明显低于基础状态组(78. 4 % ±1. 9 % , P < 0. 01) ;而AVP + IL26 组p27 蛋白表达 阳性率(60. 2 %±1. 7 %) 明显低于AVP 单独作用组(63. 3 %±1. 9 % , P < 0. 01) 。结论: IL26 能够增强AVP 诱导CFs 增殖的效应,p27 蛋白可能是IL26 和AVP 调控CFs 增殖共同的细胞内靶蛋白。
Abstract:
AIM: To study the intervention effects of interleukin-6 ( IL26) on cell proliferation and p27 expression in cardiac fibroblasts (CFs) modulated by argipressin (AVP) . METHODS: Neonatal Sprague-Dawley (SD) rats CFs were cultured as models. MTT assay was used to evaluate cell proliferation. Cell cycle distribution and p27 expression were determined with flow cytometre (FCM) . RESULTS : ①A value of CFs measured with MTT assay in the 10 000 U/ ml IL26 group was 0. 38 ±0. 01 , which was significantly higher than that of basal condition (0. 32 ±0. 01 , P < 0. 01) ; A value in 10 - 7mol/ L AVP + 10 000 U/ ml IL26 group was 0. 41 ±0. 01 , which was significantly higher than that of the AVP group (0. 39 ±0. 01 , P < 0. 01) . ②The percentage of CFs at stage S (13. 0 % ±3. 5 %) and proliferation index ( PI) (29. 4 % ±1. 9 %) in the IL-6 group were higher than those of the basal condition (7. 5 %±1. 0 % and 26. 0 %±1. 0 % , P < 0. 01) , commitment with a significant decrease in the percentage of stage G0/ G1 cells (70. 6 % ±1. 9 % vs 74. 0 % ±1. 0 % , P < 0. 01) ; the percentages of CFs at stage S (18. 8 % ±1. 5 %) and PI (35. 2 % ±1. 6 %) were much higher in the AVP + IL-6 group than in the AVP group (15. 5 % ±1. 4 % and 31. 4 % ±1. 5 % , P < 0. 01) ,commitment with a significant decrease in the percentage of stage G0/ G1 cells (64. 8 % ±1. 6 % vs 68. 6 % ± 1. 5 % , P < 0. 01) . ③Positive rate of p27 expression in IL-6 group (72. 6 % ±2. 5 %) was significantly lower than that of basal condition (78. 4 % ±1. 9 % , P < 0. 01) ; Moreover , positive rate of p27 expression in AVP + IL-6 group (60. 2 % ±1. 7 %) was significantly lower than that of AVP group (63. 3 % ±1. 9 % , P < 0. 01) . CONCLUSION: IL-6 can enhance CFs proliferation induced by AVP. p27 may be the same target regulatory protein of IL-6 and AVP in CFs.
