摘要:
目的 研究大剂量硝酸甘油(NG)对心肌缺血再灌注(MI/R)损伤的影响。方法 SD大鼠30只,制备离体MI/R模型,并将其随机分为2组:NG组和对照组。再灌注开始时NG组给予硝酸甘油(15 μg/h, 溶剂为950 ml/L乙醇, 加药速度50 μl/h),对照组给予950 ml/L乙醇(50 μl/h),观察心功能指标,检测冠脉流出液中的肌酸激酶(CK)和乳酸脱氢酶(LDH)含量,用伊文氏兰-红四氮唑(TTC)染色和TUNAL法观测再灌注心肌坏死和凋亡程度。结果 大剂量NG抑制了再灌注后心功能的恢复,引起CK、LDH释放增加,促进心肌细胞坏死和凋亡。结论 大剂量NG加重了大鼠离体MI/R损伤。
Abstract:
AIM This study examined the effects of nitroglycerin on cardiac function and cellular injury following ischemia (30 min) and reperfusion (120 min) in isolated perfused rat hearts. METHODS A large dose of nitroglycerin (nitroglycerin treated group, 15 μg/h) or 950 ml/L ethanol(vehicle, 50 μl/h)was administered at the time of reperfusion. RESULTS Administration of a large dose of nitroglycerin significantly aggravated postischemic myocardial injury characterized by depressed cardiac function recovery (P<0.05 vs vehicle), increased lactic dehydrodenase (LDH) (nitroglycerin vs vehicle: 20.3% vs 42.2%, P<0.01)and creatine kinase (CK) release (P<0.01 vs vehicle), enlarged cardiomyocyte necrosis (nitroglycerin vs vehicle: 20.3% vs 42.2%, P<0.01) and apoptosis (nitroglycerin vs vehicle: 30.2% vs 5.2 0.8, P<0.01). CONCLUSION These results suggest that a large dose of nitroglycerin may aggravate MI/R injury.
